bjective: To investigate the relationship with redox state and Yin-Yang in human blood by detecting the two pairs of redox: Glutathione of reduced/oxidized (GSH/GSSG) and coenzymeâ…¡of reduced/oxidized (NADPH/NADP+).Methods : According to Western medical and traditional Chinese medical basic standards of health, we choiced the normal adults in age 20-29,30-39,40-49,and over the age 50.Male and female proportion is fairly. All tester fasted venous blood 3ml. (1) Dithiothreitol (DTT) restores GSSG to GSH in nearly neutral conditions. GSH and fluorescence probe o-phthalaldehyde (OPA) condense isoindole which can make strong fluorescent at pH8.0. (2) The electrons of NADPH delivers to methyl thiazolyl tetrazolium (MTT) which restores to formazan under the action of phenazine methyl sulfate (PMS), while the product NADP+ can be restored to NADPH by glucose -6-- phosphate (G6P) with glucose 6 - phosphate dehydrogenase (G6PD). And then the MTT reduction as formazan. In this cycle reaction , the productivity of formazan is directly proportional with coenzymeâ…¡at different stages. Coenzymeâ…¡can be determined by the rate change of formazan absorbance.Results: (1) Male and female of the same age had no significant difference in GSH,GSSG,NADPH,NADP+,GSH/GSSG,NADPH/NADP+,Eh(GSH / GSSG),and Eh(NADPH/NADP+) (P> 0.05).(2) The was no significant difference of 20-29 and 30-39 age groups in GSH,GSSG,NADPH,NADP+,GSH/GSSG, NADPH/NADP+,Eh(GSH / GSSG),and Eh(NADPH/NADP+) (P> 0.05); 40-49 and over 50 age groups'GSH, NADPH was significantly lower than the previous two age groups (P <0.05), and over the age 50 group was lower compared with the age 40-49 ; GSSG,NADP+,GSH/GSSG,NADPH/NADP+,Eh(GSH / GSSG),and Eh(NADPH/NADP+) was significantly higher than the first two age groups (P <0.05), and over the age 50 group was higher compared with the age 40-49.Conclusion: (1)In this study,the methods of determination of fluorescence had been improved to detect GSH in the plasma more quickly and accurately. (2) The relationship of Yin-yang balance and the balance of redox state is closely. The people of Chinese and Western standards of healthy maintains a constant balance of redox state, no difference between males and females.(3)Before 40 years old, the balance of redox state has no change, After 40 years old, ages was increased and antioxidant capacity was decreased. Redox state balance gradually shifted to the oxidation, which might be suggested the turning point age of human body was 40 years old...
