Objective : CSNRDARRC is a amino acid sequence which consisted of Cys - Ser - Asn -Arg - Asp - Ala - Arg - Arg - Cys,It is an very important ligand peptide that can bind in bladdercancer cells. The project aims to study the affinity and binding site of the peptide CSNRDARRCin bladder cancer cells. Analysis of its degree of affinity in bladder tumor cells and the possibilitywhether CSNRDARRC is the peptide ligand in targeted therapy; Explore the subcellular bindingsite in bladder tumor cells. Provide experimental basis for the treatment of bladder cancer anddisease.Methods :①Label cell suspension with different concentrations(2ul,5ul,10ul)of FITCCSNRDARRCprobe, Observe the fluorescence intensity of tumor cells by flow cytometry;②After the tumor cells in the interference with peptide CSNRDARRC, then Label cell suspensionwith different concentrations of FITC-CSNRDARRC probe which is used in the previous,Observe the fluorescence intensity of tumor cells by flow cytometry;③For the subcellularbinding sites study in bladder tumor cell, the use of laser scanning confocal microscope directobservation is possible.Results : Flowcytometry showed that peptides CSNRDARRC bind to T24 in a concentration–dependent manner,the laser scanning confocal fluorescence microimaging system confirmed thatpeptides CSNRDARRC was located in Nucleus.Conclusion :1. Peptides CSNRDARRC have high affinity with human T24 bladder cancer cell2. Peptides CSNRDARRC have the binding sites in Nucleus for T24 bladder cancer cell...
