Background Pancreatic carcinoma is a common malignant tumor of digestive system,which is high malignant, mortality and recurrence rate.It's 5-year survival rate is less than 3%.Many signaling pathway's activation and deactivation can lead to oncogenesis. Hh-Gli signaling pathway plays a key role in the development of pancreatic carcinoma. There are three vertebrate Gli transcription factors (Gli-1, Gli-2, Gli-3) whose mRNA expression level can reflect the activity of Hh-Gli signaling pathway. Currently,small interfering RNA which blocked the nuclear transcription factor Gli-1 become a new treatment for pancreatic cancer. In this study, we use Gli-1 siRNA to transfect pancreatic cancer cells(PANC-1 cell line) and observe the proliferative and apoptotic effects of Gli-1 siRNA on PANC-1.Objective To observe the proliferative and apoptotic effects of Gli-1 siRNA on silencing pancreatic carcinoma cell PANC-1.Methods The cultured PANC-1 cells were randomly divided into experimental group negative control group and blank control group.In experimental group, Gli-1 siRNA was transfected into pancreatic carcinoma cell PANC-1 by using LipofectamineTM2000 and compared with the establishment of the negative control group and blank control group.24h after transfection,Gli-1 is examined by RT-PCR.The effect of cell proliferation was studied by MTT.Results After Gli-1 siRNA transfected PANC-1 cell line, PCR results showed that: the experimental group compared to negative control group, Gli-1 siRNA significantly reduced the expression of Gli-1 mRNA (P <0.05); the experimental group compared with the blank control group, Gli-1 siRNA significantly reduced the expression of Gli-1 mRNA (P <0.05); negative control group compared with blank control group, Gli-1 mRNA expression did not change significantly(P>0.05). MTT results showed that: the experimental group compared to negative control group, cell proliferation was inhibited (P <0.05); the experimental group compared with the black control group, cell proliferation was inhibited (P <0.05); negative control group and blank control group cells did not change significantly (P> 0.05).Conclusion The study provided that Gli-1 siRNA block the expression of Gli-1 mRNA and inhibit the translation and expression of downstream genes,thereby Gli-1 siRNA inhibit the proliferateion of PANC-1 cells and induce apoptosis.
