The expression of EGFR, HER2 and TOPOâ…¡in esophageal canceration course.The positive expression rates of EGFR and TOPOâ…¡display a improving trend fromnormal esophageal epithelium,intraepithelial neoplasia to carcinoma(P<0.005), in the abovethree groups,The positive expression rates of EGFR were 8.41%,15.89%,50.47%, andTOPOâ…¡were 3.74%,11.21%,43.93%.Furthermore,the expression showed gradually incresingwith histological grades advancing(P<0.01). there was no correlation between EGFR orTOPOâ…¡and gender,age,lymph node or distant metastasis(P>0.05).There was a positivecorrelation between EGFR and TOPOâ…¡(r蛋白1=0.410,P<0.05). Among the three groups,thepositive expression rates of HER2 were 0%, 1.87%, 1.87%. There were no significance of HER2protein expression among the three groups(P>0.05), no correlation was obtained between HER2and gender,age,the depth of invasion, histological grades or lymph node metastasis(P>0.05), norelationship between HER2 and EGFR or TOPOâ…¡. 2. The gene status of EGFR,HER2 and TOPOâ…¡detected by CISH in ESCC.In ESCC,EGFR gene amplifieation was found in 35(32.71%)cases,TOPOâ…¡was 31(28.97%)cases.EGFR,TOPOâ…¡positive was significantly correlated with histologicalgrades,and no correlation between EGFR or TOPOâ…¡and gender,age,lymph node or distantmetastasis(P>0.05).There was a positive correlation between EGFR and TOPOâ…¡( råŸºå› = 0.530,P<0.05). HER2 gene amplifieation was found in 2(1.87%)cases, and no correlation wasobtained between HER2 and gender,age,the depth of invasion, histological grades or lymph nodemetastasis(P>0.05), no relationship between HER2 and EGFR or TOPOâ…¡(råŸºå› 2= 0.23 1,P>0.05;råŸºå› 3=0.169,P>0.05).3. Comparision of EGFR,HER2 and TOPOâ…¡protein/gene status detected by IHC, CISH.In ESCC, the general coincident rate of EGFR or TOPOâ…¡detected by IHC,CISH was64.81%(35/54)and 65.95%(31/47). Amongst the cases of EGFR or TOPOâ…¡proteinoverexpression (+++) group,the concordant ratios of IHC and CISH detecting EGFR or TOPOâ…¡status were 80% and 83.33%; amongst the cases of EGFR or TOPOâ…¡protein overexpression(++) group, the concordant ratios were 58.97% and 60%; amongst the cases of EGFR orTOPOâ…¡protein expression (+/-) group, the concordant ratios were both 100%. The concordantratios of HER2 detected by IHC, CISH was 100%.4. The gene status of HER2 detected by FISH.Though FISH detected HER2 gene Status in 107 cases of ESCC, gene amplifieation wasfound in 1 case,others were no gene amplifieation,incluing 1 case detected Chromosome 17polysomy.Conclusion1. EGFR and TOPOâ…¡are closely related to the occurrence and development ofesophageal squamous cell carcinoma.their expressions all make a qualitative change in theesophageal high-grade intraepithelial neoplasia. The role of HER2 in the development ofesophageal squamous cell carcinoma is still not definite in ShanXi provice.2. IHC is a preliminary screening method by detecting EGFR and TOPOâ…¡protein.Because protein expression and gene amplification test results exsist some differences, we needto select CISH further for IHC(++~+++).3.Compared with the CISH, FISH play an improtant role in the detection of HER2 geneon Chromosome 17 polysomy.
