Objective:To analyze the alterations of serum proteins fingerprint of postmenopausal osteoporosis,comparing the serum proteins fingerprint of different level postmenopausal osteoporosis, to find the protein peak which has close relation with the germination and developing of postmenopausal osteoporosis.Methods:Using surface-enhanced laser desorption/ionization time-of-flight mass spectrometry (SELDI - TOF - MS) technology to weak cationic surface protein chip (CM10) for examination medium, detection 72 cases group and 70 cases of normal control group serum protein peak; to analyze the serum protein profiles of different degree cases. And analysis the differences of serum proteins fingerprint via statistics by the specialized Biomarker Wizard and Biomarker Pattern of Ciphergen Company to get the conclusion.Results:(1) according to comparing the serum proteins fingerprint between primary osteoporosis and normal person, I found there're 9 markedly differences ( p<0.05) on protein peak expression, in which there is difference when m/z is 2918, 2920, 5896, 6621, 6424, 2967, 3136, 3215,4592, protein peak decrease appeared on primary osteoporosis when m/z is 2967, and all the other increased;(2)there are differences also appeared when m/z is 9716, 9346, 4952, 5896 by comparing the osteopenia persons and those ones who have severe osteoporosis,all protein peaks increased; compare the statistics and find whether there is protein peak difference between osteopenia persons and those ones who have severe osteoporosis. But there is when m/z is 4592, 5896;(3) we can divide the normal persons and those who're osteoporosis by getting the model made by protein peak whose Molecular Weight is 2918, 5896. And the accuracy is 97.8%; Confirmed by Double blind method, the accuracy, specificity and sensitivity is 92.5%, 90%, 95%.Conclusion:There are 9 markedly differences serum protein peaks between cases and normal persons when compare them by adopting SELDI-TOF-MS; by establishing a diagnosis model for osteoporosis diagnosis and screening provides a new serological methods; it's possible to take the 4 different protein between osteopenia and severe osteoporosis as the specific protein to grade the postmenopausal osteoporosis, it provides the theory foundation for screening the differences protein between osteopenia and normal persons, and discovery and diagnosis on primary osteoporosis.
