Objective To investigate the effects of sufentanil post-conditioning on the apoptosis of hepatic cell and the expression of Bcl-2 and Bax in rat liver tissue induced by intestinal ischemia-reperfusion.Methods A total of 40 adult Wistar rats,(female/male),weighting 200-250g were randomly divided into five groups(n=8):①sham operation group(S group):performed by the same way of intestinal ischemia-reperfusion group except for ligating superior mesenterlc artery.②intestinal ischemia- reperfusion group(IIR group):ligating superior mesenterlc artery by clamp and receiving saline infusion intravenously in the same regime with those in sufentanil postcondition-ing group.③post-conditioning of low- dosage sufentanil group(SP,group).④post-conditioning of middle- dosage group (SP2 group).⑤post-conditioning of high-dosage group (SP3 group). Five minutes before reperfusion receiving sufentanil infusion respectively 1μg/kg,5μg/kg和10μg/kg. Sufentanil infusion is configured to 0.5 ml and injected in five minutes by trace pump.The model of liver injury induced by intestinal ischemia-reperfusion was established by blocking the superior mesenteric artery with clap. The left hepatic lobes were sampled after reperfusion three hours. The expression of Bcl-2 and Bax protein in hepatic tissue was detected with immunohisto-chemistry and MIAS-2000. The apoptosis index (AI)was measured by terminal deoxynucleotidy transferase mediated dUTP nick and labeling (TUNEL) method.Results①The result of light microscopy:The pathological lesion were lighter in sufentanil post-conditioning than in intestinal ischemia-reperfusion group significantly.②AI were significantly lower in sufentanil post-conditioning group than in intestinal ischemia-reperfusion group at each point(P<0.05).③Bcl-2 protein optical density was significantly higher in sufentanil post-conditioning group than that in intestinal ischemia-reperfusion group at corresponding point(P<0.05), while Bax protein optical density were significantly lower in sufentanil post-conditioning group than in intestinal ischemia-reperfusion group at each time point(P<0.05).④These indexes were not showed apparent differences among group SP1, SP2 and SP:, (P>0.05).Conclusion①Sufentanil post-conditioning can protect liver injury induced by intestinal ischemia-reperfusion in rats.②Different density was not showed different effects.③One of mechanisms of protection is may be that Sufentanil increase the expression of Bcl-2 protein and decrease the expression of Bax protein.
