ObjectivesAnalyze neurobehavioral changes of coke oven workers and their influence factors, andfurther discuss the neurotoxicity of coke oven emissions and PAH. Analyze the PBL Hsp70expression levels of coke oven workers and the relationship between single nucleotidepolymorphisms of HSP70 genes and neurobehavioral damages, and discuss the feasibility ofHsp70 expression level and HSP70 gene as a susceptible marker for neurobehavioral harm.Methods198 workers from a coking plant were selected as an exposed group (furnace bottom 23,furnace side 85, furnace roof 90), 145 anti-air staffs who were non-coking workers were chosenas a matched group. Personal information is collected by questionnaires. B [a] P and urinary1-OHP concentration in working environment was tested with the method of HPLC. Personalinformation was gathered by questionnaire. Affective state and neurobehavioral function ofrespondent was measured by NCTB/WHO. Hsp70 antigen and antibody level, lymphocyteHsp70 and single nucleotide polymorphisms of HSP70 genes in blood plasma was testedrespectively by ELISA, Western-blot and PCR-RFLP.ResultsPart 1B[a]P concentration in the air of matched group, furnace bottom, furnace side and furnaceroof was 0.01±0.00, 0.02±0.01, 0.05±0.005 and 0.21±0.02μg/m3 separately. Urinary 1-OHPconcentration of workers was 1.62±1.91, 1.86±2.19, 3.80±2.45, and 10.47±3.72umol/molCrrespectively.The test results of neurobehavior indicate that, the score difference of furnace roof workers'12 behaviors including POMST, POMSA, POMSV, POMSC, SRT, DSPF, DSPB, DSP, DSY,BVR, PAC and PA compared with the matched group, had statistical significance(P<0.05).The score difference of furnace side workers'8 behaviors including POMSA, POMSV, DSPF,DSPB, DSP, DSY, PAC and PA compared with the matched group, had statisticalsignificance(P<0.05). The score difference of furnace bottom workers'SRT compared with thematched group had statistical significance (P<0.05). Grouped by urinary 1-OHP (the grouping point is 30% and 70% of urinary 1-OHPconcentration), the respondents are classified into three groups which are low, moderate and highexposed group. The score difference of respondents'affective state among exposed groups hadno statistical significance (P>0.05), but 4 scores of POMST, POMSD, POMSA and POMSC haddeclining tendency with the increase of urinary 1-OHP concentration. The score difference ofhighly exposed group's DSPF, DSP, DSY and BVR compared with those of lowly exposedgroup and moderately exposed group, had statistical significance (P<0.05). The score differenceof each index between moderately exposed group and lowly exposed group had no statisticalsignificance (P>0.05).MLR displayed that, urinary 1-OHP level had something to do with score of emotion indexincluding POMST, POMSV and POMSC, and neurobehavioral function index including SRT,DSPF, DSPB, DSP, DSY and PA, the score of POMSV was positive correlation while the restwas negative correlation. Ages were negatively correlated with emotion scores of POMST,POMSD and POMSA, while working ages were positively correlated with them. Working agecould be introduced to DSPF and DSP regression equation and it was negatively correlated withthem. Except two emotion index including POMSF and POMSV, education degree could beintroduced to the regression equation of other index and positively correlated with their scores.Smoking index was negatively correlated with the score of POMSA, DSY and PA. Drinking wasnegatively correlated with the score of POMST, POMSD, POMSC, SRT, SRTF and DSY.Before test, job type had something to do with each affective state index except POMSD, DSYand PA, and it was negatively correlated with the score of POMST, POMSF, POMSV, POMSC,DSY and PA while positively correlated with the score of POMSA.Par 2Hsp70 expression level in lymphocyte of coke oven workers had the tendency of rise firstlyand drop later with the increase of outer exposure level. When furnace bottom group andmatched group, furnace side group and matched group, furnace side group and furnace bottomgroup are compared, the difference of Hsp70 level had no statistical significance (P>0.05).Furnace roof group and matched group, furnace bottom group and furnace side group wascompared, Hsp70 level difference had statistical significance (P<0.05).The respondents are classified by low, moderate and high expression group according totrisection range of Gamma Ratio for PBL Hsp70 expression level. People with score ofneurobehavioral test less than 50 were treated as positive neurobehavior while those with scoremore than 50 were negative. The positive ratio of respondents indicates a sequence of Hsp70 lowexpression> moderate expression> high expression. Five indexes including DSPF, DSPB, DSP,PAC and PA had statistical significance (P<0.05). When positive and negative neurobehavior of lymphocyte Hsp70 expression were compared, four test indexes including DSPB, DSP, PAC andPA had statistical significance (P<0.05). Lymphocyte Hsp70 expression in exposed group hadmore obvious correlation with neurobehavior compared with that of matched group, and it waspositively correlated with the score of 6 neurobehavioral index including DSPB, DSP, SANN,DSY, PAC and PA.Every genotype of HSP70 excluding the neurobehavioral test of DSPF, DSPB, DSP andDSY index which might be sensitive to exposure, the 11, 6 and 11 neurobehavioral index scoredifference for genotype HSP70-1 G/C, genotype HSP 70-2 G/A, and genotype HSP70-hom T/Trespectively had statistical significance (P<0.05) when exposed group was compared withmatched group, serious neurobehavioral harm was caused in exposed group compared with thatof matched group.HSP70 genes main haplotype pair and neurobehavioral score analysis result showed that, inexposed group, when GAT/GAT, GAT/GGT and GAT/CGT were compared, the SRTS scoredifference of neurobehavioral test had statistical significance (P<0.05); When GAT/GAT andGAC/CGT were compared, the score difference of 5 indexes including SRTS, DSPF,DSPB,DSPand DSY had statistical significance (P<0.05); When GAT/GAT and GGT/CGT were compared,score difference of 4 indexes including SRTS, DSPF, DSP and DSY had statistical significance(P<0.05); When GAT/GAT and GAC/GAC were compared, the score difference of 3 indexesincluding POMSV, SANP and SANN had statistical significance (P<0.05); When GAT/GATand others were compared, the score difference of 4 indexes including SRTS, DSPF, DSP andDSY had statistical significance (P<0.05). The score for people with GAT/GAT haplotype pairwas obviously lower. In matched group, the score difference for GAT/GAT haplotype pair wasonly found in PA of GAC/CGT and DSPF of others, which had statistical significance (P<0.05).The score was obviously higher than that of the above two haplotype pairs. In exposed group,when GAC/GAC was compared with GAT/GGT, the score difference of neurobehavioral testincluding POMST, POMSD, POMSA, POMSV, SANP, SANN, DSY had statistical significance(P<0.05); When GAC/GAC was compared with GAC/CGT, the score difference ofneurobehavioral test including POMST, POMSD, POMSA, POMSV, POMSC, DSPF, DSPB,DSP, SANP, SANN, DSY and BVR had statistical significance (P<0.05); When GAC/GAC wascompared with GGT/CGT, the score difference of neurobehavioral test including POMSA,POMSV, DSPF, SANP, SANN, DSY, PAC and PA had statistical significance (P<0.05); WhenGAC/GAC was compared with GAT/GAT, score difference of neurobehavioral test includingPOMSV, SANP and SANN had statistical significance (P<0.05); When GAC/GAC wascompared with Others, score difference of neurobehavioral test including POMST, POMSD,POMSA, POMSV, DSPF, SANP, SANN and DSY had statistical significance (P<0.05). The score for people with GAC/GAC haplotype pair was obviously lower. In matched group, scoredifference for GAC/GAC haplotype pair was only found in SRT of GAT/GGT and PA ofGAC/CGT (P<0.05), and the score was obviously higher than that of the above two haplotypepairs.Conclusions1. Neurobehavioral damages of coke oven workers might be caused by workingenvironment, of which the manifestation had something to do with urinary 1-OHP level.Learning and memory function including short-term listening memory, visual memory/kinematicvelocity, short-term visual memory were easily to be damaged by PAH. The neurobehavioralchanges of coke oven workers were influenced by urinary 1-OHP level, age, working age,smoking, drinking, education degree, night shift and so on.2. PBL Hsp70 expression was prohibited by high exposure level during coke oven workperiod; PBL Hsp70 expression of coke oven workers had something to do with neurobehavior.3. Single nucleotide polymorphisms of HSP70 genes of coke oven workers were correlatedwith neurobehavior to some degree. Genotype HSP70-1G/C, HSP70-2G/A, and HSP70-hom T/Twere related to neurobehavioral damages of coke oven workers under coke oven environment.Containing GAT/GAT and GAC/GAC haplotype pairs might increase the risk ofneurobehavioral damages.
