Objective:This experiment is to approach the effect of topiramate to blood magnesium and bonemetabolism of the epilepsy children, using the change of these indicatrix causing by takingtopiramate prior and post treatment. On the one hand, this experiment can evalue the security ofTPM in the children, on the other hand it can seek and estabolish a set of rational monitoringmeans and preventive approach.Method: This experiment choose 60 children with epilepsy as the experimental group (boy 32cases,girl 28 cases)and 40 healthy taking no drug as controls. The experimental group wasdivided into two groups: Single drug group(TPM only, 40 children) and Combined drug group(combination of TPM with Calcium supplement and Vitamin D, 20 children). Epilepsyclassification is to accord with the diagnostic criteria of International League Against Epilepsy in1981 and 2001.Measuring index:assessment parameters included analysis of blood magnesium, bloodcalcium,blood fosforus,BMD and determine BAP reflecting osteoplastic process .The changesof the two experimental group were observed in three different time, before TPM, 3 months afterTPM and 6 months after TPM. Then analyze the data with SPSS13.0 statistically, expressingindexes by mean, standard deviation and compareing in pairs by test.Results: The blood magnesium and bone metabolism index were not statistically significantbetween the control group and the experimental group before using TPM (P>0.05).In the Single drug group, taking TPM 6 months later, the blood calcium and BMD obviouslyreduce, they were statistically significan between taking TPM 6 months later and beforetreatment (P<0.05). However, other experimentai indexes were not statistically significantamong three stage (P>0.05).In the experimental group of combination of TPM with Calcium supplement and Vitamin D,all measuring indexes were not statistically significant between before treatment and 6 monthsafter treatment (P>0.05).Conclusion: TPM can influence the bone formation, inducing the decrease of blood calcium andBMD. However taking TPM was with no effect to blood magnesium. And the epilepsy childrentaking TPM need to be supplied with appropriate calcium supplement and Vitamin D. Thistherapeutic method can prevent and treat the adverse reactions of topiramate to bone metabolism.
