Expression Of CD4~+ CD25~+ CD127~- Regulatory T Cells In Peripheral Blood In Children With Schonlein-henoch Purpura And Its Significance

Objective:To detect the expression levels of CD4~+CD25~+CD127~-regulatory T cells in peripheralblood in children with Henoch-Schonlein purpura(HSP) in pre-treatment and post-treatmentperiod,and to investigate their significance and pathgenesis.Methods:1. experimental object:56 children with HSP before treatment,56 children with HSPafter treatment and 20 healthy children ,each one was drawn 1 ml peripheral blood,anticoagulated.2.Index detection:â‘ plus antibody: test tube is appended withCD4-PC5,CD25-FITC,CD127~-PE monoclonal antibody each 5ul,then each test tube appendedwith 10ul anicoagulated complete blood, mixed, away from light 30 minutes.â‘¡Hemolysis:Eachtest tube is appended with 1 ul hemolysin, mixed, ecaded from light 15 minutes,then centrifugedfor 5 minutes, by 1200 r/min, deserting the upper liquid.â‘¢Ablution: Putting NS 1 ml inabove-mentioned residue liquid, mixed,centrifug;ed(ditto), deserting the upper liquid.â‘£Awaitingto be detected: Putting NS 0.5ml in above-mentioned residue liquid, awaiting to be detected.Wechoose 10000 cells from each semple,anddetecting the expression of CD4~+CD25~+andCD4~+CD25~+CD127~- by flow cytometry CELLQUGST.Results: the expression of CD4~+CD25~+Treg,CD4~+CD25~+CD127~-Treg in HSP group beforetreatment was evidently lower than that in treated HSP group and control (p < 0. 01) , but therewas no remarkable difference between the treated HSP group and the control group (p>0. 05).Conclusion: the expression of CD4~+CD25~+CD127~-regulatory T cell in the treatment HSP groupwas evidently higher than untreatment HSP group,which explained that the regulatory T cellpossibly participated in pathophysiology of HSP and the development of the disease . Theexpression level of CD4 ~+ CD25~+CD127~- T cells can be used as an effective index to judge theHSP prognosis.
