Objective To research the Changes on rat heart function and myocardial structure of non-beating heart of warm ischemia caused by hyperpotassemia in order to conform the warm-ischemia time limit of the donor heart.Methods Thirty-two wistar rats were divided into 4 groups randomly:The control group (A,n=8,WI[Warm-Ischemia]=2min);The5mingroup(B,n=8,WI=5min);TheWI10mingroup(C,n=8, WI=10min), The WI 15 min group (D,n=8,WI=15min).After the WI, Langendorff Model was settled, resuscitation of the hearts were observed, the coronary flow was measured. The intra-left ventricular pressure was record and hemodynamic values were calculated. The samples of heart were observed though light microscope.Results The resuscitation was better in group A and B than in group C and D. The hearts in group A and B got sinus rhythms as soon as reperfusion initiated while all hearts in group D ventricular fibrillated (VF). Five out of eight hearts in group D could have a sinus rhythm 15 to 20 min after reperfusion started while in this group kept three VF rhythms with continuous compression. There are five hearts in group C got sinus rhythms as soon as reperfusion initiated with occasionally Ventricular premature beat (VPB) while the other hearts in group C ventricular fibrillated, two of the VF hearts in group C got sinus rhythms with compression while a heart kept VF rhythm with continuous compression. The difference of A,B,C,D four groups of hemodynamic have statistical variance, there are no significant difference between group A and group B(P>0.05),the result of hemodynamic in group C and D were significant lower than group A(P<0.05).there are no statistical variance between group C and group D(P>0.05).Conclusion warm-ischemia at least ten minutes of rat heart caused by hyperpotassemia is unfavorable for transplantation. |
