Objective:To investigate the expression of RSK4 in four human breast cell lines MDA-MB-231,T47D,Bcap-37,MCF-7,and detect their invasion and migration;Observe the relationship between the expression of tumor suppressor RSK4 gene and the invasiveness of breast cancer cell lines in vitro in order to explore the influence of RSK4 in the biological behavior of breast cancer.Methord : 1.The expression of RSK4 mRNA was detected by RT-PCR(reversetranscriptase polymerase chainreaction)in MDA-MB-231,T47D,Bcap-37,MCF-7 human breast cancer cell lines.2.The protein level of RSK4 was detected by Western-blot in MDA-MB-231,T47D,Bcap-37,MCF-7 human breast cancer cell lines;compared and analyzed the expression between Western-blot and RT-PCR methord,filtered out the most highest and lowest expression of RSK4 cells.3.The cells migration and invasion abilities were measured by Transwell chamber;analyzed the relationship between the cells migration ability and RSK4 gene.Results:1.The expression of RSK4 mRNA was the most highest in MCF-7(0.816±0.014),and decreased in Bcap37(0.683±0.030) and T47D (0.445±0.024)respectively,the expression in MDA-MB-231(0.280±0.008)was the most lowest; the statistical difference was found in pairwise comparison between the four groups (P<0.001)2.The expression of RSK4 protein level was most highest in MCF-7(0.850±0.068),and decreased in Bcap37(0.621±0.056) and T47D (0.412±0.091) respectively,the expression in MDA-MB-231 (0.222±0.030) was the most lowest, the statistical difference was found in pairwise comparison between the four groups (P<0.001)3,The expression of RSK4 mRNA in four human breast cancer cell groups presented significantly positively related to the expression of RSK4 protein level(r=0.766,P<0.001)4,The four cell groups invasiveness and migration were:MDA-MB-231(52±6) (94±5); T47D(36±3)(82±3);Bcap-37(27±2)(71±2);MCF-7(18±3)(24±4),the statistical difference was found in pairwise comparison between the four groups of invasiveness and migration(P<0.001),meanwhile,the invasiveness of the four groups pointed out significantly positively related to the migration(r=0.822,P<0.001)5,The data suggested that the invasiveness of four breast cancer cells degraded gradually following the increase of the expression level of RSK4 mRNA,it presented significant negative relation (r=-0.948,P<0.001) ; meanwhile, the invasiveness of four breast cancer cells degraded gradually following the increase of the expression level of RSK4 protein, it presented significant negative relation(r=--0.679,P<0.001)Conclusion:1,The expression of RSK4 in four different human breast cancer cell lines was: MDA-MB-231T47D>Bcap-37>MCF-7;the invasiveness and migratiopresented high concordance,both of them can stand for the cells sport ability ivitro.4,The expression of RSK4 mRNA and RSK4 protein presented significant negative relation with the invasion of human breast cancer,it accounted for that RSK4 might play some roles in the development of breast cancer , and was related with partial biological behavior . Determination of RSK4 expression level was of great value in evaluating the motility and invasiveness of breast cancer. |
