Objective:Study from the cellular level and Magnolia Combination Polygala by vitro gastric interstitial cells of Cajal(GICC) growth in mouse and its related active substances.Methods:①By Koh'method, we improved the culture of GICC, using MTT colorimetric test (MTT) to study the control group, group Polygala, Magnolia Group, Polygala and Magnolia 1:1,1:2,1:3compatibility group of GICC to optimal ratio.②Using Colorimetric, we compared the control group, Polygala serum group, Magnolia serum group, Polygala and Magnolia 1:2compatibility serum group of GICC with Lactate dehydrogenase (LDH), and calcium (Ca2+) activity of mechanism.③By the Pi method, we studied the drug-containing serum on GICC of ATPase(ATP enzyme).④Using thiobarbituric acid (TBA) method, we studied the drug-containing serum group on GICC of malondialdehyde (MDA).⑤By Griess method, we studied the drug-containing serum of GICC medium of nitric oxide (NO) levels, and investigated effect in different groups of GICC.Results:①Decoction of direct administration:Polygala group suckling mouse significantly inhibited the growth of GICC (P<0.01), Magnolia Group, Polygala and Magnolia 1:1,1:2,1:3compatibility group can significant for GICC growth (P<0.01); compared with Polygala group of 3mg/ml, Polygala and Magnolia 1:1,1:2,1:3 compatibility group can be very significant for GICC growth (P<0.01).②containing serum administration:comparison with the blank serum group, Polygala serum had the trend to inhibit the growth of GICC; compared with Polygala serum group, Polygala and Magnolia 1:1,1:2,1:3combined serum could significantly promote the growth of GICC (P<0.01).③LDH determination results showed that Polygala serum group have reduced LDH activity trends; compared with Polygala serum group, Magnolia serum group, Polygala and Magnolia 1:2compatibility group can significantly enhance the serum LDH activity(P<0.05).④The results about Ca2+ determination suggested that Polygala, Magnolia, Polygala and Magnolia 1:2 compatibility group and blank serum group of Ca2+ content show no difference (P> 0.05).⑤ATP enzymes showed that Polygala, Magnolia, Polygala and Magnolia 1:2compatibility group and blank serum group of ATPase was no significant difference (P> 0.05), but activity of Polygala and Magnolia 1:2compatibility group tended to increase slightly.⑥TBA showed that every group of MDA had no significant effect (P> 0.05).⑦the results of Griess showed different groups of GICC had no effect on NO levels (P> 0.05).Conclusion:①Polygala decoction, Polygala containing serum can significantly inhibit the growth of GICC, showing cytotoxic; It may be one of the major factors that Polygala inhibits the gastrointestinal motility.②Magnolia decoction and its containing serum group are able to promote the growth of mouse'GICC.③Polygala and Magnolia compatibility of decoction group and its containing serum group can promote the growth of GICC and increase their survival time, which may be associated with increased LDH activity. Above all, Magnolia can significantly lower inhibition of Polygala on GICC'growth, Polygala combined with Magnolia can reduce side effects of Polygala.
