Objective:Observing parallely the anti-acute brain ischemia effect of four aromatic resuscitation drugs (musk, borneol, storax and benzoin), and probing into part of its mechanism of action, and exploring the effect of acute brain ischemia on model animal heart, with the purpose of clarifying the effects of resuscitation and awakening of this kind of drug and the scientific connotation of the drug property system of "heart—bitter—floating" and the purpose of providing certain experimental basis for the initial establishment of heat stroke syndrom like animal model.Methods:①By adopting the model experiment of head breaking and mouth opening of mice, model experiment of using saturated magnesium chloride to cause acute brain ischemia of mice, and model experiment of using thread embolism method to block the middle cerebral artery of rats for 6h, which causes focal brain ischemia of rats, observing the survival time of mice, nervous function of model rats, area of cerebral infarction, and change in brain tissue morphology and evaluate the anti-acute brain ischemia effect of aromatic resuscitation drug.②Measuring the activity of superoxide dismutase (SOD), content of malondialdehyde (MDA), vitality of Na+-K+-ATPase and Ca++-Mg++-ATPase, and content of serum nitric oxide (NO) in the brain of model rats with focal brain ischemia, and probing into part of the action mechanism of anti- brain ischemia of aromatic resuscitation drug.③Observing the electrocardiogram and changes in cardiac muscle tissue morphology, activity of heart SOD, content of MDA and vitality of Na+-K+-ATPase and Ca++-Mg++-ATPase of model rats with focal brain ischemia by thread embolism method, and exploing the correlation between brain ischemia and heart function.④Measuring the changes in body temperature, heart rate and blood pressure of model rats to provide certain experimental basis for the duplication of animal model of "heat stroke syndrom".Results:①Aromatic resuscitation drug represented by musk and borneol could increase the times and time of mouth opening of mice whose head had been broken, and could prolong the time of mouth opening for breathing of mice injected with saturated magnesium chlorid solution (P<0.05).②After the middle cerebral artery of rats had been blocked by the thread embolism method, the time for coming around of rats in model group were prolonged (P<0.05). These rats closed their eyes tightly and the adducted forepaws at the opposite side of nidus could not stretch completely, displaying damages to nervous function (P<0.05), and their body temperature increased significantly (P<0.05).③Compared with the model group, aromatic resuscitation drug represented by musk and borneol could reduce the damage to nervous function of rats with focal brain ischemia caused by thread embolism method (P<0.05), the area of infarction by focal brain ischemia of mice and rats and pathological changes (P<0.05).④Aromatic resuscitation drug represented by musk and borneol could reduce the MDA content in the brain of rats with focal brain ischemia by thread embolism method (P<0.05), raise the activity of brain SOD (P<0.05), and increase the vitality of Na+-K+-ATPase and Ca++-Mg++-ATPase in the brain of model rats with brain ischemia (P<0.05), but had no influence on the content of serum NO (P>0.05).⑤Compared with the sham operation group, no pathological change had happened to electrocardiogram and cardiac muscle tissue morphology of rats with focal brain ischemia for 6h by thread embolism method, and no significant change had taken place in the activity of cardiac muscle SOD, MDA content, and vitality of Na+-K+-ATPase and Ca++-Mg++-ATPase (P>0.05).Conclusion:①Aromatic resuscitation drug represented by musk and borneol has the effect of anti- acute brain ischemia, the action mechanism may be correlated with the reduction of the brain MDA content and the increase of brain SOD activity and the vitality of Na+-K+-ATPase and Ca++-Mg++-ATPase in the brain of rats with brain ischemia.②Model of "heat blocking syndrom" like rats has been established. The differences in the effects of the four aromatic resuscitation drugs on the temperature, heart rate and blood pressure of model animal have reflected the differences of this kind of drug in the "4 properties". The anti-acute brain ischemia effect of aromatic resuscitation drug is the surface feature of its drug effect of resuscitation and awakening, and is also the drug effect spectrum of the "bitter—heart—floating" property system.④For rats with focal brain ischemia for 6h by thread embolism method, no pathological change has been found to the brain.
