Pertactin is an important component of acellular pertussis vaccine. Natural PRN has disadvantages in purification process. Because of low cost and easy purification, recombinant PRN was considered as a good substitute in acellular pertussis vaccine. In this pAPVer, we evaluated the immunogenicity, antigenicity and protection of recombinant PRN.The PRN antibody was detected after two immunizations of recombinant PRN and natural PRN and showed no significant difference between them (p>0.05).The recombinant PRN had good immunogenicity and antigenicity in our study.After immunized with recombinant PRN the mice were efficaciously protected from the infection of pertussis 18323. Among high, medium and low immunization doses, the medium dose showed the highest protection rate. The survival rate of high dose in two analog vaccines(recombinant PRN+vaccine stock solution+FIM2+FIM3,natural PRN+vaccine stock solution+FIM2+FIM3) was the same, which is 93.75%. While middle-dose and low-dose survival rates in recombinant PRN group were higher than the control group.The recombinant PRN group could secrete not only lower levels of Thl type cytokines, such as IL-2, IL-10, IFN-γ, but also higher levels of Th2 type cytokines, such as IL-4 and IL-12,which implied that the recombinant PRN can stimulate humoral immunity and cellular immunity, and mainly the humoral immune response. The type of immune response of the natural PRN group is the same as the recombinant PRN group. Two analog vaccines (r-PRN+vaccine stock solution+FIM2+FIM3 and n-PRN+vaccine stock solution+FIM2+FIM3) also stimulated humoral immune response and cellular immunity. Thus, they led to a polarization of the immune response toward the Th2 type.Both IgGl and IgG2a could be secreted by recombinant PRN group and nature PRN group. It indicated that each group can stimulate the organism to produce a certain degree humoral immunity and cellular immunity. Thus, they led to a polarization of the immune response toward the Th2 type.This study indicated that the recombinant PRN will be a good candidate to replace the natural PRN APVplyling to pertussis vaccine development.
