Object:to explore the level of traditional Chinese medicine Panax notoginseng saponins (PNS) for the treatment of Alzheimer's disease and its part of the role of its possible mechanism from animal,designed to combat Alzheimer's PNS to provide experimental and theoretical basis.Methods:Select the 2-month-old SD female rats were 15 for the young control group,while 22-month-old SD female rats were 45 which are randomly divided into old control group,low-dose group and high-dose drug group. Application of Y-maze test is used in rats to test the learning and memory function, the evaluation index is the correct reaction times and correct response on rates. The hippocampus CAI area of the histological is graded with light microscope, neuronal density is calculated with high-powered microscope.TUNEL is used to test hippocampal CAI neuronal apoptosis. Results:1)the correct number of times in Y-maze learning test, each group of three times in the training results,the 2nd correct number is more than the first one second,3rd times is more than the first two; the Old model group at the same time significantly is less than the correct number of young control group (P<0.01);low-dose group at the same time correct number is more than old model group,(P<0.05).High-dose group at the same time correct number is more than old model group, P<0.01).2)Y-maze test properly of correct response rate:Aged model group learning and memory performance was significantly lower than the young control group, P<0.01;PNS treatment group can obviously improve the learning and memory function in aged rats,compared with the old model significant difference, P<0.01; High-dose group of learning and memory performance is better than the low-dose group, P<0.05, which reflect the PNS to improve learning and memory function in rats with dose-related.3) The histological grade hippocampus CAI area and neuronal density showed that the young control group of histological grade is 0-1 grade, neuronal density is 197±10.1, while the aged model group of histological grade is 2-3 level, neuronal density is 66±13.5, compared with the young group there was significant, P<0.01.PNS of low-dose group of the hippocampus histological grade is 1-3 level, neuronal density is 101±12.4,compared with the old group was significant, P<0.05.High-dose group hippocampus histological grade is 1-2 level. Neuronal density is 149±14.3, compared with the old group was significant,P< 0.05.4)the number of TUNEL-positive cells in the hippocampal CAI area:the number of neuronal apoptosis of aged model group is more than the young control group, PNS low-dose group and PNS high-dose group, significant difference is existed,P<0.01, to take the youth group at least, followed by high-dose group;the low dose group and the comparison of high-dose group showed high-dose group were obviously less neuronal apoptosis, P<0.05.Conclusion:Panax notoginseng saponins can improve learning and memory ability in aged rats to reduce rat brain tissue,particularly the hippocampus tissue damage.PNS effects are dose-response relationship,the role of high dose is stronger than low-dose.
