Objective Many people had studied on the dissection physiology,pathology of cardiac lymph in 1960. Miller found cardiac intimal fibrosis was the important mechanism of blocking cardiac lymph flow in 1963. Recently people found blocking cardiac lymph flow was correlated with coronary artery disease and myocardium interstitial fibrosis.Liu found there were endothelium cell swelling,array disorder,media edema and fibroblast proliferation on coronary artery and small arterys wall in10~14day after blocking goat cardiac lymph flow.Fibrosis proliferation were observed in adventitia and myocardium interstitial in 60~180 day.He thought the reason of fibrosis proliferation was a great deal of protein stimulated fibroblast to proliferate. The aim of this study was discuss the mechanism of myocardium interstitial fibrosis after blocking lymph of rabbit heart by investigating the change of plasma C-terminal propeptide of typeⅠprocollagen (PICP),N-terminal propeptide of typeⅢprocollagen (PIIINP) and angiotensinⅡ(AngⅡ) and measuring the express of collagenⅠ,ⅢmRNA.Metheods 34 New Zealand white rabbits were divided into experiment group and control group randomly. Animals were anesthetized with intravenous sodium pentobarbitone (30 mg/kg). The animals were artificially ventilated with room air at the rate of 60 strokes/min. The thorax was performed in median incision and the cardiac was suspended in a pericardial cradle.0.5 ml of 10% methylen blue was injected into the left and right ventricle near the apex of the heart. Methylen blue stained and clearly marked the lymphatic vessels in the epicardium. The main lymphatic vessels seen on the epicardium were all ligated. The large lymph nodes between the aortic root and pulmonary artery, and between the posterior aorta and the right superior pulmonary artery were also destroyed with electric cautery. The chest was then closed by layers and the animals were returned to the animal house for standard care by the investigators and the animal house curators. Assaying PICP,PIIINP and AngⅡbefore operation and at 3,7,14,30,60 days after operation by Enzyme-linked immunoadsordent assay(ELISA).Expression of collagenⅠ,ⅢmRNA were measured in situ hybridzation pre-operation and 7,14,30,60 days.Result The concentration of plasma PICP,PIIINP and AngⅡof group E have no significant difference between 3,60 days and pre-operation(P>0.05);There was no significant difference group E and group C(P>0.05). The plasma PICP,PIIINP and AngⅡof group E at 7,14,30 days were higher than pre-operation and the group C (P<0.05).There was no significant difference between pre-operation and after operation in group C;Plasma PICP,PIIINP were correlated with AngⅡ;Expression of collagenⅠ,ⅢmRNA of group E were consistent with Plasma PICP,PIIINP and AngⅡ.Conclusion Plasma PICP,PIIINP and AngⅡbegin to increase from 7 days after operation until 30 days and be normal at 60 days. Increaseing of collagenⅠ,Ⅲwas one of reasons of myocardium interstitial fibrosis resulted from blocking lymph.
