Objective:To invest the reparation role and the possible mechanism of transpl-anting Human umbilical cord masnchymal stem cells to ATNMethods:1.Modelling Method and Grouping:(1)Fourteen healthy dogs were randomly divided into two groups.Model group 1:Each dog were given 0.2%HgCl2,7mg/kg by intramuscular injection to establish model of Acute tubular necrosis.By peripheral veins injection,transplanted Human Umbilical cord Mesenchymal Stem Cells that were cultured in vitro and identified by DAPI.Model group 2:Modelling Method was the same as Model group 1;(2)The rats were divided into three groaps.â‘ ATN with cells transplantation groap:inject HUMSCs into caudal vein at the 3rd day of gentamicin injection.â‘¡ATN groap:inject gentamicin for 3 days only.â‘¢control group:the normal rats without any intervention.Made an ATN model induced by gentamicin:injected a total dose of 900mg/kg gentamicin subcutaneously within 3 days.2.Cell Transplantation:(1)Then by femoral artery intervention, transplanted Human Umbilical cord Mesenchymal Stem Cells cultured in vitro and identified by DAPI into the left renal artery.(2)To transplant into SDrats from the tial.3.Index test:(1)24h later,killed all dogs and took kidney organization to observe nephridial tissue by light microscope and the survival of Human Umbilical cord Mesenchymal Stem Cells by fluorescence microscope.(2)â‘ Biochemical indicators detection:urine protein,BUN,SCr,â‘¡Kidney pathological section:observed the renal pathologic change by HE,â‘¢Traced the HUMSCs homing:under the wave of 370nm ultraviolet light with fluorescence microscope.â‘£Detected the BMP-7 and Bcl-2 expression levels in kiney by Immunohistochemistry.Results: 1.The transplated Human Umbilical cord Mesenchymal Stem Cells can survive in the of Model group 1 and Model group 2.There was little differences on fluorescence intensity between two groups (P>0.05).2.A total dose of 900mg/kg within 3 days could make model successfully,A large first dose could reduce the mortality rate.3.In cell transplantation group,the rats'hairs grew burnish,the mortality rate lowered obviously.4.Such indexes as BUN,SCr,and etc.arrived peak value at 1w in cell transplantation group and ATN group,but higher in front of the three time points in ATN group.5.To observate renal tubular epithelial cells In ordinary light microscope6.Observed transplant group renal tubular parts show the green fluorescence, while the glomerular area were negative.7.BMP-7and Bcl-2 expressed abundantly on 1d in cell transplant-ation group and reached its peak on 4d.Conclution:1.Human Umbilical cord Mesenchymal Stem Cells can survive in the kidneys of ATN and there is no obvious difference on treatment effect between peripheral veins injection and femoral artery intervention.2.HUCMSCs can treat Acute tubular necrosis effectively3.HUCMSCs can enhanced the expression of Bcl-2 in the kidneys,alleviated the apoptosis of renal tubular injury and increased renal tubular epithelial cell proliferation.As a result,renal tubular and renal function was restored.4.HUCMSCs can reduced the expression of BMP-7 in the kidneys to provide an appropriate micro-environment for the local kidney tissue regeneration.
