PurposeSal B (SalB), a multi-phenolic compounds, has the potential ability to affect the heart, it is the heart of the protection mechanisms involved in anti-oxidation and regulation of MMP-9 activity. However, anti-oxidation and regulation of MMP-9 activity in the basic mechanism of SalB is not so clear, this article is to study whether SalB can antioxidant and regulate MMP-9 activity.Methods1. The Research uses isolated heart model attempts to explain one of the mechanisms to protect the heart. The rats are randomly divided into four groups:model group (control) and of three administration groups of SalB (15,30, and 60μmol/L), the isolated heart using Langendorff device, pre-perfusion 25min, occlusion 30min, reperfusion 40min. Hemodynamic parameters were recorded, included:LVEDP, LVDP,±dP/dtmax,. Experimental determinated the creatine kinase (CK) vitality; Then At the end of Experiment, malondialdehyde(MDA),superoxidi dismutase(SOD) were measured by enzymatic method.2. the rats are randomly divided into four groups:sham operation group (Sham), sham operation + Sal B group (Sham + SalB), operation group (AMI), operation group + Sal B group (AMI + SalB), respectively, producing myocardial ischemia model, the model uses the Miller catheter measured hemodynamic data; ligation after 24 hours, the rats were killed, open the chest, the heart cut out, leaving the ventricle, for TTC staining; part of the heart to stay for histopathological study; an alternative determination of tissue lesions of the content in MMP-9.Results1. Compared with control group, there are phenomenon in three dose groups that LVEDP dropped, while the LVDP,+dP/dtmin,-dP/dtmin are rising. Note after administration, isolated rat heart hemodynamics improved. Biochemical measurements showed that the treatment group can reduce the CK (creatine phosphate enzyme) activity and MDA content, increased SOD activity.2.In the TTC staining experiments, the administration significantly reduced the percentage of stained area; AMI-SalB group histopathological hit shows sparse arrangement of myocardial cells, stripes and clearer than the model group, myocardial interstitial edema, without inflammatory cell infiltration. AMI group compared with a marked improvement; and In the determination of MMP-9 levels, SalB is inhibiting its growth, compared with AMI group, obviously statistically significant. ConclusionFully described above, SalB resistant to oxidative stress and regulation of MMP-9,The research can expound the protection of a possible new mechanism of SalB in ischemic injury rat heart. It is not only To deepen the study of Salvia, but also for clinical application to provide a new theoretical basis.
