Background and purposeGastrointestinal stromal tumors (gastrointestinal stromal tumor, GIST) as a new concept which was put forth in the 20th century,80 years, with the continuous development of molecular biology techniques and progress of studies have shown that, GIST has become the most common mesenchymal tumors in the digestive tract currently, GIST as a non-directional differentiation of gastrointestinal stromal tumors, the purpose is to distinguish it from occurring in the gastrointestinal tract and other myogenic or neurogenic sarcoma, due to lack of specific immunohistochemistry in the past, electron microscopy alone morphologically difficult to differentiate tumor types, such tumors are often misdiagnosed as the source of smooth muscle or nerve tumors. At present, although there is no judge of a unified standard of benign and malignant GIST, but most people think that tumor size and mitotic gastrointestinal stromal tumors such as an important prognostic indicators for the same time, whether the tumor is ulcerated also have an important impact on prognosis. At present, with regard to GIST research and Zoological Cytological studies have reported less, this paper in the establishment of subcutaneous tumor model in nude mice under the premise of more individuals in different-week-old nude mice subcutaneously transplanted tumor growth in different parts of the situation for the future other diseases, tumor-bearing nude mouse model for the establishment of a reference, and in vivo to study gastrointestinal stromal tumor cells, the occurrence, development and a variety of biological characteristics and a variety of anti-cancer drug screening provides an indispensable tool; for build a platform for further experimental study.Materials and methodsAccording to the 40-week-old nude mice were randomly divided into two groups and then to vaccination sites were randomly divided into two groups, a total of four groups, to take fresh stromal tumor surgical specimens, sterile conditions, divided into 2cm * 2cm * 2cm size of the tissue block,. Would be a good separation of stromal tumor tissue in nude mice were inoculated at 4 weeks of age, under the arms, buttocks four-week-old nude mice,8-week-old nude mice under the anns,8-week-old nude buttocks area will be kept in specific pathogen free (SPF) environment, the constant temperature (22-26℃), humidity (40-60%) environment, once every three days recording the volume of tumor growth until the tumor grew to 1cm3, and draw the growth curve, in nude mice were sacrificed and observed tumor morphology, visceral metastasis, and parallel histopathological and immunohistochemical detection of gastrointestinal stromal tumors and human samples can be compared; measurement data in each group using x±s said that the application of fourfold table x2 testing, analysis of variance of data processing, p<0.05 indicated statistically significant differences, SAS software for statistical analysis.Results1. tumor formation rate:The tumor was inoculated into 4-week-old nude armpit area (100%) than in nude mice inoculated at 4-week-old hip (80%) subcutaneous tumor formation rate is high; inoculated 4-week-old nude armpit site (100%) was higher than the armpit of nude mice inoculated at 8 weeks of age, location (70%) into the tumor formation rate is high; inoculated 4-week-old nude buttocks area (80%) than in nude mice inoculated at 8 weeks of age, the hip area (40%), tumor formation rate is high; above p<0.05, the difference was statistically significant. But the tumor was inoculated into 8-week-old nude armpit area (70%) and 8-week-old nude mice were inoculated into the hip area (40%), p> 0.05, still can not explain the difference between statistical significance.2. ulceration rate:tumor of nude mice inoculated at 4 weeks of age, the hip area (10%) was higher than the armpit of nude mice inoculated at 4 weeks of age, position (0%) ulceration rates; inoculated 8-week-old nude buttocks area (20%) was higher than the armpit of nude mice inoculated at 8 weeks of age, position (0%) ulceration of the high rate, the above p<0.05, significant difference; while tumors in nude mice inoculated 8 weeks of age, the hip area (20%) and inoculation site at 4-week-old nude buttocks (10%) compared, p> 0.05, still can not explain the difference between statistical significance, and the 4-week-old nude mice inoculated in the armpit and the armpit area of the eight-week-old nude mice tumors were not ulcerated phenomenon (0%).3. softening disappearance rate:The tumor was inoculated into 4-week-old nude buttocks (10%) was higher than the armpit of nude mice inoculated at 4-week-old site (0%) softening rates; inoculated 8-week-old nude armpit (30%) than in the armpit of nude mice inoculated at 4 weeks of age, position (0%) softening of the high rate; inoculated 8-week-old nude buttocks area (40%) than in nude mice inoculated at 4 weeks of age, hip (10%) softening of the high rate of, the above p<0.05, significant difference; while tumors in nude mice inoculated 8-week-old buttocks (40%) than in the armpit of nude mice inoculated at 8 weeks of age (30%), both of softening rate is relatively, p> 0.05, still can not explain the difference between statistical significance.4. rate of tumor growth:tumor of nude mice inoculated at 4 weeks of age, the armpit area (21.00±2.00 days) than the armpit of nude mice inoculated at 8 weeks of age (27.86±1.46 days) required for a short time; inoculated at 4 weeks of age, nude buttocks (25.33±1.58 days) than the 8-week-old nude mice were inoculated buttocks (31.50±3.00 days) required for a short time, the above p<0.05, significant difference; while tumors in nude mice inoculated 4-week-old axillary Under the site with the 4-week-old nude mice inoculated tumor hips comparison; 8-week-old nude mice inoculated armpit area and buttocks of nude mice inoculated at 8 weeks of age, tumor site compared to the growth rate difference was not statistically significant (p> 0.05) ConclusionIn this study, different ages in different parts of nude mice xenograft situation, and so a comparison, draw the following conclusions:1. Nude-week-old smaller the higher the rate of specimen into the tumor, and the inoculation site in the armpit into the tumor site compared with the high rate of hip; weeks of age, the greater the tumor formation rate, the lower, and inoculated in different parts of the specimen into the tumor There was no significant difference.2. Regardless of the size-week-old nude mice inoculated in the buttock area greater than the rate of tumor ulceration ulceration of the rate of axillary part of the tumor, but the ulceration rate and the size-week-old nude mice was no significant difference. The study also found that:the larger-week-old nude mice, regardless of how the tumor inoculation site, the tumor disappeared on their own higher the rate of softening, but in smaller-week-old nude body, the hip is higher than the rate of tumor disappeared on their own armpit transplant site.3. Regardless of how the vaccination site, the smaller-week-old nude mice, tumor growth faster; but for the same week-old nude mice, regardless of how the vaccination site, tumor growth rate were not significant difference.GIST in nude mice the growth of the comparative study for the future state of in vivo studies of gastrointestinal stromal tumor cells, the occurrence, development and a variety of biological characteristics and a variety of anti-cancer drug screening provides an indispensable tools; for other tumor diseases, the establishment of tumor-bearing model provides a reference for further experimental study to build a platform.
