1.1 ExtractionAs a Chinese traditional medicine, Saururus chinensis (Lour) Bail, is a perennial herbaceous plant, whose whole plant can be used as a medicine in traditional Chinese medicine. The main chemical composition of Saururus chinensis (Lour) Bail contains lignans, flavonolds, alkaloids, tannins polysaccharide, as well as amio acids have been discovered. harmacological studies have shown that Saururus chinensis (Lour) Bail have clearing Heat and expelling Miasma,against hydropsy and other effect.Profound research has been made of chemical composition of Saururus chinensis by this paper. We got nine compounds from the plant, and seven of their chemical stuctrues were identified by the methods of IR,MS NMR and so on.The seven compounds'names are Sauchinone, Licarin B, Licarin A, Saucernetin, Aristolactam BⅡ, Aristolactam BⅡand Saurloactam, respectively.Besides, this experiment also established two lignans'simultaneous deteemination method by HPLC.Dried herb samples (9Kg) were crushed to powder and refluxed three times by 85% ethanol (the volume fraction). Every time,the volume of ethanol used were 50L. And the three extraction time were 3h,2h and 1h. Combined extract, recovery solvent to no alcohol flavor, diluted with water and then passed through AB-8 macroporous adsorption resin. AB-8 macroporous adsorption resin was eluted by the volume fraction of 85% ethanol. After recovering organic solvent, and 216g extraction was obtained.1.2 Isolation purification and IdentificationAfter isolating from the extraction by Silica-gel, ODS column chromatography and other methods, nine compounds were obtained.Through the Physico-Chemical Properties and their characters of UV,MS, 1D-NMR, the seven compounds'chemical structures were identified,and their names are Sauchinone, Licarin B, Licarin A, Saucernetin, Aristolactam BⅡ, Aristolactam BⅡand Saurloactam.2 The Contents determination of Sauchinone and Saucernetin2.1 Equipment, reagents and reference substanceChromatograph:Agilent 1200 HPLCDetector:PDWD variable wavelength UV detectorColumn:ZORBAX SB C18 coloum(250mm×4.6mm,5μm)Detection wavelength:232nmBalance:Mettlertoledo AG135 electronic balanceReagents:Chromatography of methanol,distilled water twiceReference substance:Sauchinone(compound G-1,98.8%) and Saucernetin(compound G-9,99.0%)Mobile phase:methanol-water gradient elutionThe methanol set A and the water set B,0-8min:0%-50%A,8-20min:50%-60%A,20-25min:60%-65%A2.2 Methodology validation1)The linear relationshipIn the range of 0.2~20μg, the peak area(Y) as the vertical axis, the injection mass (X,μg) as the abscissa,the linear regression equations of G-1 and G-4 were: Y=2.083×103X-117.63 r=0.9998 Y=0.973×103X+55.224 r=0,99982) Detection limitsWhen the concentration of sample was 2×10-4μg, the S/N of the two compounds both were between 3-4,so the detection limit was 0.1μg.3)Precisiones The same sample was tested 6 times in parallel.The RSD of peak area was 1.31%(G-1) and 0.46%(G-1).The RSD of in parallel. time was 0.02%(G-4)and 0.04%(G-4).4)RepeatabilitiesIn the 6 copies of the same sample solution,G-1 and G-4 content of the RSD values were 1.55% and 1.86%, respectively.StabilitiesThe same sample was tested once every 2 hours within 12 hours.The RSD of the peak area of compound G-1 was 1.36%; RSD of the peak area of compound G-4 was0.71%.The results showed that compound G-1 and G-4 was stable within 12 hours and the ready sample solution can be determined within 12 hours.6) RecoveriesThe average recovery of compound G-1 was 100.3% and the RSD was 0.68%;The two values of compound G-4 were 101.7% and 1.53%.2.3 DeterminationThe average content of the compound G-1 was 0.050% and Compound G-4 was 0.024%.In this paper, the chemical constituents of Saururus chinensis (Lour) Bail..was furtherly studied, and new index of content determination was offered to improve quality standard of the plant.The results of this paper provided a theoretical support for further study and a scientific basis for development and utilization of Saururus chinensis (Lour) Bail..
