Objective:To explore the role of SRC-3 and MMP-9 in the development of adenomyosis and provide the experimental data and theoretical basis for clinical diagnosis and treatment of adenomyosis. Methods:1. Experiment samples:42 cases of ectopic endometrium were collected in this study and 10 cases of control endometrium were obtained from the area of non-tumor in hysterectomy uterus with uterine fibroids.2. Indicators and methods:All of the patients had not been treated with hormone and anti-adenomyosis medication six months before operation. The pathological diagnoses were confirmed by two pathologists. The expression of SRC-3 and MMP-9 were measured by immunohistochemistry and tissue microarray. Image was analyzed by Image Pro 6.0.3.Statistical analysis: The statistical methods were carried out by the SPSS 17.0 software, and the significant level was defined as p<0.05. Results:1. The expressions of SRC-3 and MMP-9 in ectopic endometrium were significantly higher than that in control endometrium(p<0.01); 2. (1)The expressions of SRC-3 and MMP-9 in control endometrium at proliferative phase were significantly higher than that in secretory phase(p<0.05). (2)The differences of the expressions of SRC-3 and MMP-9 in ectopic endometrium between proliferative phase and secretory phase were not significant (p>0.05), which were not related to menstrual cycle.3. The expressions of SRC-3 and MMP-9 in the ectopic endometrium during proliferative phase and secretory phase were significantly higher than that of concurrent control endometrium(p< 0.05); 4. (1) The expressions of SRC-3 and MMP-9 had no significant correlation in control endometrium (r=0.401,p=0.196>0.05). (2) The expressions of SRC-3 and MMP-9 had a significant positive correlation in ectopic endometrium (r=0.720, p=0.008<0.05).Conclusion:1. The expression level of SRC-3 in the ectopic endometrium of adenomyosis was significantly higher than that in control endometrium. Overexpressed SRC-3 might make the endometrial cells proliferate by improving of estrogen sensitivity. Consequently, ectopic implantation and growth could be achieved easily.2. The overexpressions of MMP-9 in the ectopic endometrium might implicate that MMP-9 can enhance the degradation of extracellular matrix.3. The overexpressions of SRC-3 correlated to positive of MMP-9 in the course of adenomyosis, which might play an important role in the development of the adenomyosis,but the specific mechanism remains to be further studied.
