Objective: LPS and smoked to replicate rat model of COPD, By weight and pathological changes of lung tissue to evaluate the role of prevention and treatment of Astragalus injection; By the diversification of IL-8 and IL-10 in serum and BALf to investigate the mechanism of the role of prevention and treatment of Astragalus injection in rat model of COPD Methods: (1)Sixty SPF level male rats were divided into four groups randomly: Normal control group 10 rats,model group 20 rats,early intervention group 15 rats, late intervention group 15 rats. On the two points of day 1and day 14,To the model group, the early intervention group and the late intervention group were injected with LPS intratracheal 200μg/μl,the rats were placed in the remaining days of the smoke chamber in the fume. On the two points of day 1and day 14, To normal control group were injected equal volume of saline, And fed in normal air conditions, 28 days. Biopsy to determine a successful model for relication(2) In the day 31,all the rats were killed, Take right middle lobe to make paraffin sections, By radiommuoassay and ELISA,four groups were measured in serum,bronchial lavage fluid IL-8, IL-10 levels, and for the BALF cell count. Results: (1)COPD rat model confirmed successfully by pathological reproduced. (2) The role of prevention and treatment: weight loss of model group compared with the normal group were satatistically significan(tp<0.05,p<0.01),weight gain of the early intervention group and the late intervention group compared with the model group were satatistically significant(p<0.05,p<0.01); By HE staining of lung tissue, We found lung injury of early intervention group and late intervention group were more significantly reduced than the model group. (3) the mechanism of the role of prevention and treatment of Astragalus injection in rat model of COPD. serum IL-8 content: the content of IL-8 was 2.05±0.43ng/ml in model group, the normal group was 0.78±0.19ng/ml,compared with normal group , model group were significantly different(p<0.01). the content of IL-8 was 0.73±0.17ng/ml in early intervention group,the late intervention group was 0.70±0.18ng/ml , compared with model group, early intervention group and late intervention group were satatistically significant(p<0.01).BALF IL-8 content: the content of IL-8 was 0.49±0.20ng/ml in model group than the normal group 0.29±0.16ng/ml was significantly higher(p<0.01). the content of IL-8 was 0.35±0.17ng/ml in early intervention group, the content of IL-8 was 0.13±0.09ng/ml in late intervention group, compared with model group were satatistically significant(p < 0.01).the late intervention group than the early intervention group decreased more significantly IL-8(p<0.01). serum IL-10 content: the content of IL-10 was 129.08±92.69 pg/ml in model group, the normal group was 256.53±64.63pg/ml,compared with normal group , model group were significantly lower(p < 0.01). the content of IL-10 was 141.79±66.29pg/ml in early intervention group, the content of IL-10 was197.07±43.27pg/ml in late intervention group, compared with model group were satatistically significant(p<0.05,p<0.01). the late intervention group than the early intervention group raised more significantly IL-10(p<0.01). BALF IL-10 content: the content of IL-10 was 10.54±26.28pg/ml in model group, the normal group was 148.14±45.48pg/ml,compared with normal group , model group were significantly lower(p<0.01). the content of IL-10 was 42.57±46.56pg/ml in early intervention group, the content of IL-10 was 75.05±76.03pg/ml in late intervention group, compared with model group were satatistically significant(p < 0.01). the late intervention group than the early intervention group raised more significantly IL-10(p<0.01);cell count of BALF: the white blood cell count, neutrophil count , neutrophil percentage of model group compared with normal group were significantly higher (p<0.01).,compared with early intervention group and late intervention group were significantly lower (p<0.05),the two intervention groups, the difference was not statistically significant(p>0.05)Conclusion: (1) COPD rat model confirmed successfully(2) Astragalus can increase the body weight of rats COPD,can improve pathological changes of lung tissue,the late intervention group was better than the late intervention group in the role of prevention and treatment in rat model of Chronic Obstructive Pulmonary Disease . (3) The mechanism of the role of prevention and treatment of Astragalus injection in rat model of COPD were associated with the decrease of IL-8 and the increase of in serum and BALF .
