ObjectiveObjective: To study the influence on the receptor of advanced glycation end-products andligand-S100,HMGB1 by OM antagonize SiO2 stimulating the alveolar macrophages. Methods:SD neonatal rats'lung fibroblasts were purify-cultivated and stimulate-cultivated. These cellswere divided into five groups: the blank control group; the 100μg/ml SiO2 group;the 100μg/mlSiO2+200μg/ml OM group; the 100μg/ml SiO2+400μg/ml OM group; the 100μg/mlSiO2+800μg/ml OM group,the blank control group adding eagles solution without fetal calfserum. Four parallel samples were designed for each group. These cells were cultured for 4hours,24 hours and 48 hours.the RAGE was observed by means of immunohistochemicalfluorescent cells technique and Western blot; the S100 was observed by means ofimmunohistochemical fluorescent cells technique;ELISA was adopted to detect the level ofHMGB1, TNF-αand TGF-βin the supernatants of alveolar macrophages. Results:1.The resultsof the RAGE showed: at the same time,the expression of RAGE increase with the increasingconcentration of OM,and in addition to the 48h 100μg/ml SiO2+800μg/ml OM group, the otherexpression of RAGE in each experimental group are lower than the blank control group,thediscrepancy is significant (P<0.05);at the different time,in the different concentration group ofOM,with the extension of time,the expression of RAGE increase, the discrepancy issignificant (P<0.05).but in the group of SiO2,with the extension of time,the expression of RAGEdecrease,the discrepancy is significant (P<0.05). 2. The results of the S100 showed: at the sametime,the expression of S100 decrease with the increasing concentration of OM.,and in additionto the 48h 100μg/ml SiO2+800μg/ml OM group, the other expression of S100 in eachexperimental group are higher than the blank control group,the discrepancy issignificant (P<0.05);at the different time,in the different concentration group of OM,with theextension of time,the expression of S100 decrease, the discrepancy is significant (P<0.05). but inthe group of SiO2,with the extension of time, the expression of S100 increase,the discrepancy issignificant (P<0.05). 3. The results of the HMGB1 showed: at the same time,the expression ofHMGB1 decrease with the increasing concentration of OM,and in addition to the 24h and 48h100μg/ml SiO2+800μg/ml OM group, the other expression of HMGB1 in each experimentalgroup are higher than the blank control group,the discrepancy is significant (P<0.05);at thedifferent time,in the different concentration group of OM,with the extension of time,theexpression of HMGB1 decrease, the discrepancy is significant (P<0.05). but in the group ofSiO2,with the extension of time, the expression of HMGB1 increase,the discrepancy issignificant (P<0.05). 4. The results of the TNF-αshowed: at the same time,the expression ofTNF-αdecrease with the increasing concentration of OM,and in addition to the 48h 100μg/mlSiO2+800μg/ml OM group, the other expression of TNF-αin each experimental group are higherthan the blank control group,the discrepancy is significant (P<0.05);at the different time,in thedifferent concentration group of OM,with the extension of time,the expression of TNF-αdecrease, the discrepancy is significant (P<0.05). but in the group of SiO2,with the extension of time, the expression of TNF-αincrease,the discrepancy is significant (P<0.05). 5. The results ofthe TGF-βshowed: at the same time,the expression of TGF-βdecrease with the increasingconcentration of OM.,and in addition to the 48h 100μg/ml SiO2+800μg/ml OM group, the otherexpression of TGF-βin each experimental group are higher than the blank control group,thediscrepancy is significant (P<0.05);at the different time,in the different concentration group ofOM,with the extension of time,the expression of TGF-βdecrease, the discrepancy issignificant (P<0.05). but in the group of SiO2,with the extension of time, the expression ofTGF-βincrease,the discrepancy is significant (P<0.05). ConclusionConclusion:1. RAGE express in thenormal alveolar macrophage, SiO2 can reduce its expression. 2. SiO2 promote alveolarmacrophage secrete S100, HMGB1, TNF-αand TGF-β, and with the extension of reaction time,the secretion increased.3. OM can antagonize SiO2 stimulate alveolar macrophages on theimpact of RAGE and ligands.
