| Objective: To study the mitochondrial DNA D-loop region instability in acute leukemia andlymphoma.Methods: The HV-1 region and HV-2 region of D-loop region of 30 acute leukemia andlymphoma patients were analyzed by PCR-SSCP,then amplificated , sequenced and comparedwith revised Cambridge reference sequence(rCRS),mtDB and oral epithelium of the samepatien,then The mutation rates was measured by SPSS11.5 statistics software.Results: 1.In the acute leukemia group:(1) The total mutation rate was 66.67%(20/30) of our patients.There were 59 mutationsin 25 genes,with a new gen mutation: nt16209T>C; and four instable microsatellites of D310(nt303-315C7TC5),D16184(nt10184-10193C5TC4),nt203-208(GT)3, nt16388-16391 G4. Thegen mutations involved by all patients in common prognosis group werent150C>T,D310C7TC5(C8TC6) > C7TC6 or C9TC5,D16184C5TC4>C10 or C11. The gen mutationsinvolved by all patients in bad prognosis group were nt152T>C,nt16311 T>C,nt16519 T>C.(2) The mutation rates of the untreated group,PR group and CR group wererespectively 2.19×10-3±0.07×10-3,4.37×10-3±1.83×10-3 and 2.87×10-3±0.81×10-3.One-wayANOVA resulted in F=5.421,P=0.015<0.05,further analysis with LSD-t test revealed thePUT-PR=0.005<0.05,PPR-CR=0.031<0.05 and PUT-CR=0.299>0.05.(3) The mutation rate comparison between the common prognosis group and the badprognosis group: the mutation rate was respectively 3.01±1.24×10-3 and 3.55±1.73×10-3,t=-0.825,p=0.420>0.05.(4)The mutation rate is not correlative with sex and age.2. In the lymphoma group:(1)The total mutation rate was 46.67%(14/30) of our patients.There were 54 mutationsin 31 genes, with eight instable microsatellites of D310(nt303-315C7TC5),D16184(nt10184-10193C5TC4),nt 16259-16262 C4,nt 16388-16391 G4,nt 16470-16474G5,nt203-208(GT)3(,nt 233-241(TAA)3,nt 353-356 C4. (2) The mutation rates of the untreated group,PR group and CR group were respectively3.28×10-3±1.55×10-3,6.12×10-3±2.52×10-3 and 3.28×10-3×10-3±0.77×10-3.One-way ANOVAresulted in F=4.093,P=0.047<0.05,further analysis with LSD-t test revealed thePUT-PR=0.037<0.05,PPR-CR=0.028<0.05,PUT-CR=1.00>0.05.(3) The mutation rate comparison between the common prognosis group and therefractory prognosis group: the mutation rate was respectively 4.26×10-3±1.82×10-3 and4.37×10-3±3.22×10-3, t=0.82,p=0.936>0.05.(4)The mutation rate is not correlative with sex and age.ConclusionsConclusions: 1.There was a high mutation rate and were various typies of mutations ofmitochondrial D-loop region in acute leukemia and lymphoma,which led to a doughty instability.2.The chemotherapy could cause a more doughty D-loop region instability.3. The mutation rate of D-loop region maybe decrease with the release of theacute leukemia and lymphoma and the reduction of chemotherapeutics.4.The gen mutations which maybe correlative with AL prognosis werent150C>T,D310C7TC5(C8TC6) > C7TC6 or C9TC5,D16184C5TC4>C10 or C11, nt152T>C,nt16311T>C,nt16519 T>C. |
