The Expression And Its Significance Of HGF And TGF-β₁ In Obstructive Nephropathy In Rats

Objective To study the expression of HGF and TGF-β1 in renal interstitial fibrosis of complete unilateral ureteral obstruction rats and its significance in renal interstitial fibrosis.Methods 48 Wistar rats, 24 control group, according to the experimental group after 1 week, 2 weeks, 4 weeks divided into 3 groups, n = 8. Line abdominal incision the left side of the skin, muscle, and abdominal wall layers, storm drain and separated the left ureter, ureteral ligation in UPJ, Department to establish CUUO model. Semi-quantitative reverse transcription polymerase chain reaction and immunohistochemical assay (1 week, 2 weeks, 4 weeks) and compensatory obstructed kidneys and sham operation group HGF and TGF-β1 expression. According to the results of the experimental groups for the correlation analysis.Result CUUO model and the compensatory obstructed kidneys HGFmRNA and HGF protein expression in ureteral obstruction after 1 week, 2 weeks, 4 weeks at different time points were significantly higher than the kidneys (P <0.05). As the obstruction time, HGFmRNA (P <0.05) and HGF protein (P <0.05) increased. And compensation for the same subjects is much higher than that of the obstructed side HGFmRNA (P <0.05), and HGF protein (P <0.05). CUUO model of TGF-β1mRNA and TGF-β1 protein expression in ureteral obstruction after1 week , 2 weeks 4 weeks at different time points was significantly higher than the control group. No significant differences between the compensatory side and the control side. Ureteral obstruction in 2 weeks between HGF and TGF-β1 showed a negative linear correlation (r =- 0.943, p <0.05).Conclusion With the severity of the renal interstitial fibrosis, the expression of HGF in the remnant kidney tissues gradually increased, suggesting that HGF may play an important role in the pathogenesis of renal fibrosis. HGF and TGF-β1 showed a negative linear correlation in two weeks, suggesting that renal interstitial fibrosis these two factors have a close relationship to the renal interstitial fibrosis, or by inhibiting the expression of TGF-β1 and enhance the expression of HGF may enhance prevention and treatment of renal interstitial fibrosis provides a new idea.
