Objectice: In this study, immunohistochemistry was used to investigate expression situation of bFGF and uPA protein in different clinical TNM stages of nasopharyngeal carcinoma (NPC) specimen, analyzing its relationship with clinical pathological factors and the survival time after patients receiving treatment. The purpose was to study their roles in invasion and metastasis of NPC and to provide new theoretical foundation for forecasting the early diagnosis, prevention and clinical treatment, judging prognosis of NPC.Methods: 1. Samples of complete clinical data 155 cases of NPC and 20 cases of nasopharyngeal mucosa with chronic inflammation tissue were detected by SupervisionTM two-step method of IHC for bFGF and uPA proteins expression. 2. SPSS13.0 statistical software was used for statistic analysis. A p-value less than 0.05 was considered significance. The correlation between the expression of bFGF and uPA and major clinical pathological characteristics of NPC were analyzed by Chi-square test and Spearman analysis. Post-treatment survival period analysis was computed by the Kaplan-Meier method and compared by Log-rank test.Results: 1.The positive expression rate of bFGF and uPA in NPC specimens was 80.6% and 79.4 %, respectively. Positive rate of bFGF and uPA expression in nasopharyngeal mucosa chronic inflammation tissue was no expression. There was a statistically significant difference. 2.The expression of bFGF and uPA proteins was not correlated with gender, age. 3. Postitive expression rate of bFGF and uPA in the clinical TNM stages ,T1,T2,T3,T4 respectively 53.3%,75.9%,84.2%,96.6% and 53.3%,66.7%,91.2%,93.1% ,Two-parameter comparison between the T stages there was a significant statistical significance ( P<0.01 ) ; N1,N2,N3,N4 respectively77.8%,69.8%,80.9%,94.7% and 70.4%,69.8%,82.3%,92.1%, N stages between the two indicators were significantly statistically significant(P<0.05); M0,M1 respectively 78.0%,95.7% and 76.5%,95.7%, Two-parameter comparison between the two groups were statistically significant(P<0.05).4.The positive expression rate of bFGF and uPA proteins inâ… ~â…¡,â…¢,â…£a,â…£b stages are respectively 43.3%,87.7%,88.9%,95.7% and43.3%,84.2%,88.9%,95.7%. Two indicators in the comparison between different clinical stages were prompted to have statistically significant(P<0.01). 5.There were significantly positive correlation between bFGF and uPA expression in NPC(r=0.557,P=0.000). 6. Median survival period of patients with bFGF or uPA expression singly or their combinated expression was shorter than that with bFGF or uPA negative. Patients with lymph node metastasis, distant metastasis, and later TNM stages associated unfavourable prognosis.Conclusions: 1.The positive rate of bFGF and uPA expression in NPC tissue was higher that in nasopharyngeal mucosa chronic inflammation tissue. It shows two genes may take part in the occurrence and development of NPC. 2. bFGF and uPA was not correlated with gender, age, and was related to TNM stages and clinical stages of NPC patients. The result suggested that bFGF and uPA expression were prone to local invasion, lymph node metastasis, and distant organ metastasis. 3. There was significantly positive correlation between bFGF and uPA expression in NPC. It indicated that they were likely to co-promoting factors of NPC in invasion and metastasis. 4. bFGF and uPA expression were associated with NPC patients of prognosis. bFGF and uPA positive expressed in NPC patients with shorter survival time and poorer prognosis. And their combined expression had the risk of decreasing survival time of patients with NPC.
