Dangguibeimukushen Pill's Effect On The Expression Changes Of PCNA, Bcl-2 And Bax In BPH Mice Model

Objective: To study the effect of Dangguibeimukushen Pill on benign prostatic hyperplasia (BPH) and its possible mechanism.Methods: The BPH mouse model is produced by intraperitoneal injection of testosterone propionate. Sixty male mice are randomly divided into six groups which is control group, model group, positive control group, high, medium and low dose treatment group, and each group has ten. Except the control group with the same dose of olive oil, the other five groups were injected testosterone propionate (12.5mg/ kg-1) into peritoneum to create model. The high, medium and low dose treatment group were fed with Dangguibeimukushen Pill, and the dose is 24g/kg·d^-1,12g/kg·d^-1,6g/kg·d^-1 respectively. The positive control group was fed the Qianliekang liquid 0.6g/kg. Killed this five groups mouse on day 21 to observe the prostate index changes, HE staining changes of prostate pathological observed by light microscopy and the expression changes of PCNA, Bcl-2 and Bax detected by immunohistochemistry.Results: Compared with model group, the prostate wet weight and the prostate index of control group is lower than the model group(P<0.05); the PCNA, Bcl-2 expression of control group measured by IOD values is lower than the model group(P<0.05); the Bax expression of control group is higher than the average IOD value of the model group(P<0.05); the prostate wet weight and the prostate index of treatment group is lower than the model group(P<0.05);he PCNA, Bcl-2 expression of treatment group measured by IOD values is lower than the model group(P<0.05),and the Bax expression of high and meduim treatment group is higher than the average IOD value of the model group(P<0.05).Moreover, there is no significant difference between the treatment groups in prostate wet weight, the prostate index and the expression of PCNA,Bcl-2,Bax(P>0.05),and also the difference between treatment groups and positive control group in prostate wet weight, the prostate index and the expression of PCNA,Bcl-2 and Bax has no statistical meaning(P>0.05).Conclusion: Dangguibeimukushen pill can suppress experimental mice BPH; the mechanism may be related to the reduction expression of cell-proliferation protein PCNA and inhibitor of apoptosis protein Bcl-2, to the increased expression of Bax.
