With the people living standard rise ceaselessly, the water quality problem has attracted more and more attention, the sanitary quality problem caused by microbial contamination is also a concern. According to the estimate of world health organization, during billions patients of food-borne diseases in the world every year,70 percent have eaten food and water contaminated by a variety of pathogenic microorganisms.E.coli is the largest number of bacteria in human and animal intestinal tract, and most of them are non-pathogenic, but the pathogenic Escherichia coli invade some parts of the body, can cause diarrhea, cholecystitis, cystitis, etc. People who are infected with pathogenic Escherichia coli have some symptoms of pain in the stomach, vomiting and fever, the consequences might even be fatal when the baby and the old man are infected.E.coli is the international hygiene indexes which is generally used in water quality, it commonly used to instruct the feces polluted condition in order to control water quality and keep safety. Therefore, establishing a new rapid detection method of E.coli, has important significance of improving environmental health and the quality of people's life.The details are listed below:(â… )Through OD value measurement and plate counting, the 13 substances sorted to 4 kinds bacterial growth stimulants were studied on the growth of E.coli, The selection of rapid growth substances will provide a theoretical basis for the improvement of E. coli rapid detection culture medium. (â…¡) The Escherichia coli Chromogenic media was put into the different concentrations of sucrose, monopotassium phosphate and tween-20 to improve the detection rate. The Escherichia coli were detected by improved Escherichia coli Chromogenic media in different water.And compared with the MTF method, the average detection rate is 11.2% higher than MTF.(III) Applying Tanon colony counting systems to count automatically, by this way can it reduce counting time. Escherichia coli in urban sewage, Suzhou river, Jingyue lake, were detected by Tanon colony counting systems, and the automatic counting is compared with artificial counting, and it has some advantages such as count quickly high automaticity and widely used.(IV) A new method of detecting E. coli is set up, it is based on the relationship between the microbe initial concentration and the culture time of exponential phase, it also provides a new idea for the research of E. coli rapid detection.
