Infectious diseases, whether influenza, common cold, infectious mumps, polio, measles, herpes keratitis, hepatitis, SARS, such as or the latest H1N1, most are caused by specific viruses. In viral infection, the most common respiratory virus infection. Currently, the clinical use of antibiotics and antiviral drugs are mostly synthetic drugs, toxic side effects of these drugs after discontinuation recurrence rate is high, resistance. Thus efforts to find effective antibacterial,antiviral activity and low toxicity of Chinese medicine is very important. In recent years, a large number of clinical observations and experimental results show that Trollius has a good antibacterial and antiviral for the treatment of respiratory tract infection. However, its mechanism is unclear, limiting its clinical application. Currently, the active ingredient of Chinese medicine Trollius, most documents are represented by flavonoid compounds, including the identification of crude drugs and medicines for the quality assessment also are indicators of flavonoids, which are mainly orientin and vitexin.In order to clarify the Trollius chinensis Bunge antiviral activity and mechanism of this nasturtium in on anti-viral active ingredients flavonoids were extracted, purified; and its main active ingredient orientin were isolated and identified. At the same time using UV spectrophotometry and HPLC method for the determination of flavonoids and Trollius orientin content and purity, the flavonoid content of 15.42%, orientin content was 1.161% , purity 98.12%。In vitro cell culture techniques, use common respiratory tract infections caused by viruses [respiratory syncytial virus (RSV),3 influenza A virus (INF), Coxsackie virus (Cox-B3), 7 type adenovirus (Adv7)], and Hep-2 passage cells to ribavirin for the control of flavonoids, orientin had antiviral activity; and take two routes of administration to observe the cytopathic effect (CPE): the anti-virus adsorption on Direct inactivation of viruses and viral replication inhibition, to clarify the mechanism of orientin. The results show that the flavonoids and orientin inhibit RSV, INF,Cox-B3 growth, not inhibit the growth of Adv7. The mechanism of antiviral drugs and the virus while mixing, 37°C for 1 hour, plus transfer to the Hep-2 cells, the cells no CPE hole appeared,of the drug played in vitro cell culture antiviral mechanism of a drug adsorbed on the cell surface or enter the cell, to prevent virus adsorption and entry, to protect the cells.
