Objective:to discard polysaccharide from Shengjingtang, and conduct animal experiments to compare the affect in promoting ovulating of polysaccharide, Shengjingtang and Shengjingtang being discarded polysaccharide using in the rats with polycystic ovary syndrome, and to make evaluation of guiding the preparation of medicines, and further clarify the active ingredients.Methods:take 60 clean SD female rats,which are 70 days old, weighing 240g±20g.Control group which contains 10, the remaining 50 using Poresky way that take HCG (3IU/d) plus insulin (dose from 0.5IU/d to 6IU/d and maintained to the end of the modeling) subcutaneous injection for 23 days induced the formation of polycystic ovary syndrome, after modeling divide the rats into model group and treatment group 4 groups (including positive drug group, Shengjingtang group, polysaccharide group and Shengjingtang that bengjing discarded polysaccharide treatment group), n= 8, to remain 8 rats selected under the weight in the control group. Gavage with different drugs under requesting after the group were administered, once a day, for 14 days. The last day all rats were fasted overnight, decapitated for collecting blood the next day,determine the content of E2, P, T in blood. Dissected and record the weight of the uterus and ovaries, formaldehyde fixed sections of ovarian tissue to ovarian pathological observation.Results:The model rats with polycystic ovary syndrome compared with normal ones serum E2, P significantly decreased P<0.01, serum T was significantly increased P<0.05. Shengjingtang being discarded polysaccharide group serum E2, P levels increased compared with model group, serum T concentration decreased and P<0.01, significant difference;Shengjingtang being discarded polysaccharide group serum E2, P, T levels increased compared with Shengjingtang group P>0.05, no significant difference. Polysaccharide group compared with the model group serum E2, P content increased, decreased serum T, but P> 0.05, no significant difference.Uterine weight of Model group is more than the normal group, but P> 0.05, the difference was not significant. Polysaccharide group compared with the normal group after treatment uterine weight is increased, P<0.05, has significant difference. Model rats abnormal expression of ovarian weight compared with normal weight increased significantly P<0.05. Shengjingtang being discarded polysaccharide group ovarian weight compared with the model is decreaded P<0.05 significant difference; Shengjingtang being discarded polysaccharide group compared with Shengjingtang group ovarian weight is increased, P> 0.05, the difference was not significant. Ovarian weight of polysaccharide group compared with the model group P> 0.05, no significant difference.Conclusion:(1)Poresky law model features PCOS serum E2, P highly decreased, serum T significantly higher and the weight of ovarian and volume are increased. (2)Discard polysaccharide from Shengjingtang, does not affect the overall efficacy, due to the concentrated active ingredients to increase serum E2, P enhance the effectiveness, thereby enhancing the regulation of ovarian function, in order to achieve a stronger role in promoting ovulation. (3)Polysaccharide cannot increase serum E2, P and decrease the level of serum T, can not regulate the function of ovarian, but can enhance the function of uteru.
