Capillary electrophoresis is an important and powerful analytical separtion tool. The advantages of CE including small sample size, high resolution and sensitivity, nice selectivity, reduced cost and so on. It has been developed widely in recent years. Electrochemiluminescence (ECL) has many advantages including its simple instruments, low background noise, wide linear range and so on. Nowadays, the technology of CE coupled with ECL (CE-ECL) has been paid considerable attention. CE-ECL is widely used in the separation and analysis of trace components and complex samples.In this study, the CE-ECL method was applied to analyze four medicaments, which were atomoxetine hydrochloride, Lincomycin hydrochloride, Metronidazole and Ephedrine hydrochloride. The study of atomoxetine hydrochloride and ephedrine hydrochloride with three proteins was investigated. The three proteins were BSA, cytochrome C (cyt-C) and Myoglobin (Mb).The main contents are described blew:1. Through the optimization of experimental conditions, the CE-ECL analysis system has been applied to the determination of the atomoxetine hydrochloride. The optimal conditions are as follows:the detection potential at 1.15 V; 5 mmol/L Ru(bpy) 32+dissolved in 70mmol/L of pH8.0 in detection cell; the electrokinetic injection at 12 kV for10 s; 12.5 kV separation voltage; the running buffer was 30 mmol/L sodium phosphate of pH 7.5. It was successfully used for the determination of the concentration of atomoxetine hydrochloride in atomoxetine hydrochloride capsules and rat plasma. It could provide a new method of real samples detecting for atomoxetine hydrochloride.2. It was found that, the CE-ECL analysis system can meet the need for the analysis of atomoxetine hydrochloride. Based on this observation, CE-ECL was appiled to the study the interaction of atomoxetine hydrochloride with three protein, which were BSA, cytochrome C (cyt-C) and Myoglobin (Mb). And the binding constants were 9.51×103L/moL,1.15×104L/moL and 7.38×103L/moL, respectively. And the binding sites were 15.23,10.19 and 17.15, respectively.3. A method of capillary electrophoresis(CE) with tris(2,2'-bipyridyl) ruthenium (Ⅱ)electro-chemiluminescence(ECL) detection has been applied to the determination of Lincomycin hydrochloride in Lincomycin hydrochloride injection.The effects of detection potential,concentration and acidity of phosphate buffer, separation voltage, injection voltage and time, concentration and acidity of seperation buffer were investigated. Under optmimzed conditions, the linear range was 5×10-8~9×10-6g/mL, the detection limits was 1.55×10-8g/mL, the recoveries were between 99.62% and 100.31%.4. A fast and sensitive appprach of capillary eclctrophoresis(CE) with tris(2,2'-bipyridyl) ruthenium (Ⅱ) electro-chemiluminescence(ECL) detection has been applied to the determination of metronidazole in metronidazole injection.The effects of optmimzed conditions were that detection potential at 1.20 V,5 mmol/L Ru(bpy) 32+and 60 mmol/L phosphate buffer with PH 7.0 in the ECL cell, separation voltage at 15 kV, electrokimetic injection voltage at 10 kV and time for 10 s, the seperation buffer was 15 mmol/L (pH 7.0) sodium phosphate. Under optmimzed conditions, the linear range was 8×10-8~2×10-6g/mL (R2=0.997), the detection limits was 9.7×10-9g/mL. The method was successfully used for the analysis of the content of Metronidazole in Metronidazole injection samples in the two different manufacturers and in injectio metronidazoliet glucosi in one manufacturer. The recoveries were between 99.01% and 100.75%.5. CE-ECL detection of Ephedrine hydrochloride based on Ru(bpy)32+was prescented. The ECL intensity was linear with the Ephedrine hydrochloride concentration ranged from 6×10-8 to 6×10-6g/mL under the optimal conditions. The limit detection was 4.51×10-9g/mL (S/N=3). This method was successfully utilized to Ephedrine Hydrochloride the determination in Ephedrine Hydrochloride and Nitrofurazone Nasal Drops, the recoveries were ranged from 96.14% to 100.29%.6. A new method for the study the interaction of Ephedrine hydrochloride with three protein, which were BSA, cytochrome C (cyt-C) and Myoglobin (Mb) was established by CE-ECL. The binding constant were 1.551×104 L/moL,6.58×103 L/moL and 1.59×104L/moL,respectively. And the binding sites were 8.52,12.60 and 10.66 respectively.
