Objective:Osteosarcoma is a highly malignant primary bone tumor, with rapid development, significantly local failure and poor prognosis. Nowadays, many researches on osteosarcoma were mainly carried out from the mechanism of cytokines and growth factor. In contrast, researches on the relationship between osteosarcoma and the role of endocrine hormones and their receptors were rare. Former reports on calcitonin focused on multiple tumors. However, expressions of relevant researches on calcitonin receptor in osteosarcoma were seen nowhere. This experiment, primarily by immunohistochemical method, clarifies calcitonin receptor expression in osteosarcoma in order to further explore the factors of influencing the development of osteosarcoma and its prognosis.Materials and methods:1.Specimen collection:specimens were used as the experimental group of osteosarcoma and normal bone tissue as the control group. Experimental group,35 cases of osteosarcoma specimens, was divided into well-differentiated osteosarcoma group with 23 cases and poorly differentiated osteosarcoma group with 12 cases..2.Routine paraffin section.3.Immunohistochemistry:staining by SP method of immunohistochemistry4.Data processing:Contrast the ratio between positive specimens and negative specimens, based on the immunohistochemistry. x2 test or Fisher's exact test from statistical software SPSS 13.0 was used to analyze the experimental data.Results:1. Calcitonin receptor in osteosarcoma was positive, the positive rate of 68.6%; Calcitonin receptors in normal bone tissue was positive, the positive rate of 30%.2. The expression rate of Calcitonin in well-differentiated osteosarcoma was 78.2%, and the expression rate of calcitonin in poorly differentiated osteosarcoma was 33.3%. 3. The positive expression rate of CTR in male and female well-differentiated osteosarcoma patients was 77.8% and 22.2% respectively; and the negative expression rate was 60% and 40% respectively; The positive expression rate of CTR in male and female poorly differentiated osteosarcoma patients was 50% and 50% respectively; and the negative expression rate was 62.5% and 37.5% respectively。Conclusion:1. Calcitonin receptor expressed in osteosarcoma, and its quantity was higher than normal osseous tissue, the difference is significance.2. The quantity of calcitonin receptor is higher in high differentiation group than in low differentiation group, the difference is significance.3. The calcitonin receptor expression in osteosarcoma has no significant difference with Gender and the incidence of the disease.4. After calcitonin receptor combine with calcitonin, may direct restraint bone destruction of osteoclast in osteosarcoma and restraint osteosarcoma.5. To treatment osteosarcoma with calcitonin, the bone destruction of osteosarcoma can be restrainted.
