Objective:Hypoxia is the main factor to stimulate the secretion of EPO in body. In recent decades, research and practice has been fully demonstrated that high altitude training can stimulate the body's secretion of EPO, thereby to increase RBC, Hb and HCT level and enhance the body's aerobic capacity. The purpose of this study is designed by animal experiment to observe the effect of different hypoxic trainings on EPO in serum, RBC, Hb, HCT and VEGF in serum of rats, and obtain the experimental evidence that the rats have adapted to hypoxic training and the suitable mode of hypoxic training.Methods:120 male Sprague-Dawley rats of 8 weeks, which average weight is 182.8±7.3g, were divided randomly into 6 large groups:control group(C), regular aerobic exercise group(E),15% oxygen concentration control group(15C),15% oxygen concentration exercise group(15E),11% oxygen concentration control group(11C) and 11% oxygen concentration exercise group(11E)(n=20). Each large group divided into 7 days group and 21 days group (n=10). Animal in exercise group daily exercised on treadmill for 30min once a day and the speed was 12m/min.15C and 11C were placed in the hypoxic room of 15% and 11% oxygen concentration 2h every day.15E and 11E were placed in the hypoxic room of 15% and 11% oxygen concentration 2h every day, the first 30min was treadmill exercise and placed in the hypoxic room 1.5h after exercise. A total of 7 days continuous training. After 7 days' hypoxic training, derived material from 7 days group's animal. After 15 days, derived the same material from 21 days group's animal. According to the design of all experimental groups after the end of exercise programs, all rats are anaesthetized and inferior vena cava blood sampling. RBC, Hb and HCT were measured by automatic blood cell analyzer. EPO in serum were measured by American ADL goat anti rat Erythropoietin Elisa assay kit. VEGF in serum were measured by American ADL rat anti human Vascular Endothelial Growth Factor Elisa assay kit.Results:1. The effect of different mode of hypoxia on EPO in serum(1) The results showed that comparing with the control group, the concentration of EPO in serum of rats of 11% and 15% oxygen concentration control group were higher than the control group after 7 days'hypoxic exposure, but no significant difference was observed. (2) Comparing with the regular aerobic exercise group, the concentration of EPO in serum of rats of 11% and 15% oxygen concentration exercise group were higher than the regular aerobic exercise group and the 15% oxygen concentration exercise group significantly higher than the regular aerobic exercise group(P<0.05). Comparing with the 11% oxygen concentration exercise group,15% oxygen concentration exercise group were significantly higher than 11% oxygen concentration exercise group(P<0.05). (3) The number of RBC of rats2. The effect of different mode of hypoxia on RBC(1) The results showed that comparing with the control group, the number of RBC of rats of 11% and 15% oxygen concentration control group were significantly higher than the control group after 7 days'hypoxic exposure(P<0.05). (2) After 7 dats of hypoxic training, comparing with the regular aerobic exercise group, the number of RBC of rats of 11% and 15% oxygen concentration exercise group were significantly higher than the regular aerobic exercise group (P<0.05). (3) The number of RBC of rats in each group had significantly reduced after 15 days(P<0.05).3. The effect of different mode of hypoxia on Hb(1) The results showed that comparing with the control group, the concentration of Hb of rats of 11% and 15% oxygen concentration control group were significantly higher than the control group after 7 days'hypoxic exposure(P<0.05). (2) The results showed that the concentration of Hb of rats of 11% and 15% oxygen concentration control group were higher than the regular aerobic exercise group after 7 days' hypoxic exposure, but no significant difference was observed. (3) The concentration of Hb of rats in each group had reduced after 15 days. The 11% and 15% oxygen concentration exercise group had significantly reduced after 15 days(P<0.05).4. The effect of different mode of hypoxia on HCT(1) The results showed that comparing with the control group, the concentration of HCT of rats of 11% and 15% oxygen concentration control group were significantly higher than the control group after 7 days'hypoxic exposure(P<0.05). (2) Comparing with the regular aerobic exercise group, the concentration of HCT of rats of 11% and 15% oxygen concentration exercise group were significantly higher than the regular aerobic exercise group(P<0.05). (3) The concentration of HCT of rats in each group had significantly reduced after 15 days(P<0.05).5. The effect of different mode of hypoxia on VEGF in serum(1) The results showed that comparing with the control group, the concentration of VEGF in serum of rats of 11% and 15% oxygen concentration control group were lower than the control group after 7 days'hypoxic exposure and the 15% oxygen concentration control group had significantly lower than the control group(P<0.05).(2) The results showed that comparing with the regular aerobic exercise group, the concentration of VEGF in serum of rats of 11% and 15% oxygen concentration exercise group were lower than the regular aerobic exercise group after 7 days' hypoxic exposure and the 11% oxygen concentration exercise group had significantly lower than the regular aerobic exercise group(P<0.05).Conclusions: 1. Hypoxic exposure and exercise under hypoxia can enhance the concentration of EPO in serum of rats under different oxygen concentrations. The effect is the most obvious when exercises under 15% oxygen concentration.2. Hypoxic exposure and exercise under hypoxia can change the number of RBC of rats under different oxygen concentrations. The effect is the most obvious when stays under 15% oxygen concentration and staying alone is more obvious than exercising in hypoxic environment.3. Hypoxic exposure and exercise under hypoxia can change the concentration of Hb of rats under different oxygen concentrations. The effect is the most obvious when exercises under 11% oxygen concentration.4. Hypoxic exposure and exercise under hypoxia can reduce the concentration of VEGF in serum of rats under different oxygen concentrations.5. The number of RBC and the concentration of Hb and HCT can enhance after 7 days'hypoxic exposure and hypoxic training. They will reduce after 2 weeks.
