0bjective To assess the role of aquaporin 1 (AQP1) and vascular endothelial growth factor (VEGF) in the regulation of amniotic fluid volume (AFV). Methods Immunohistochemical technique was employed for detection the expressions and intensity of AQP l and VEGF on the human placenta and fetal membranes in 20 cases of oligohydramnios,22 cases of normal term pregnancy and 17 cases of polyhydramnios. Ten cases in every group using random selection were used for western blot.Results 1.Both placenta and fetal membranes yielded positive results by immunohistochemistry, AQP1 and VEGF were both localized in placental vascular endothelial cells and syncytiotrophoblasts,epithelial cells of theamnion , and cytotrophoblasts of the chorion leave .But there were no significant difference in the intensity of AQP1 and VEGF expression levels on the placenta of three groups using one-factor analysis of variance (ANOVA).Expression levels of AQP1 on the fetal membranes in oligohydramnios,normal AFV and polyhydramnios groups respectively were 5.82±1.33,6.75±1.25,7.94±1.85. Expression levels of VEGF respectively were 4.83±1.07,5.65±1.42,6.70±1.53. There were significant differences in the intensity of AQP1 and VEGF expression levels on the fetal membranes of three groups using LSD (Least Significant Difference) analysis among them.2. Expressions of AQP1,VEGF protein could be deccted in three gpoups using western blot. There were no significant differences on the placenta. Integrated optical density(IOD) of AQP1 in three groups on the fetal membranes were 13.35±3.33,16.21±2.45,19.51±2.84 respectively, IOD of VEGF were 12.24±2.12,14.37±1.90,16.51±2.09. There also were significant differences of AQP1 and VEGF IOD on the fetal membranes of three groups using LSD analysis.3. Expression levels of AQPl and VEGF were relevant to amniotic fluid volume. There were also positive correlation between AQP1 and VEGF. Conclusion The up-regulations of AQPl and VEGF protein on the fetal membranes may promote the intramembranous absorption.
