A new method for determination of ephedrine and pseudoephedrine in ephedra and maxingshigan tang by capillary zone electrophoresis(CZE) was established. In this paper, the CZE signals of maxingshigan tang were de-noised by using wavelet transform,and the change of chemical constituents of dissimilarity set square of maxingshigan tang were analyzed using wavelet toolbox.The results indicate that:1. The conditions of the experiment were optimized with a fused-silica capillary of 60 cm×50μm i.d. (50 cm effective length ) in a running buffer of 50 mmol·L-1 borax-20 mmol·L-1 threonine (pH9.27) with an applied voltage of 15 kV at room temperature. Sample introduction was performed by hydrodynamic injections (10 cm×20 s) and determined with on-column UV monitoring at 210 nm. Phenobarbital was chosen as the internal standard. Ephedrine and pseudoephedrine were separated successfully within 8 min. The linear responses covered the ranges from 21.3 to 213μg·mL-1 (r=0.9996) for ephedrine and from 8.4 to 84μg·mL-1 (r=0.9995) for pseudoephedrine. The limits of detection (S/N=3) of ephedrine and pseudoephedrine were 1.45μg·mL-1 and 1.48μg·mL-1, respectively. The limits of quantification (S/N =10) of ephedrine and pseudoephedrine were 4.81μg·mL-1 and 4.93μg·mL-1, respectively. The average recoveries for ephedrine and pseudoephedrine were 97.5% and 98.6% with RSD less than 5.0%. The method is simple, rapid, cost-effective and precise with satisfactory results.2. Due to the basic theory, uncertain enviormnental disturbance and various elecrtophoresis conditions,noise and real CE signals often coexist in the profile,which will greatly affect the accuracy and reproducibilty of the results. In this paper, the off-line CE signals were deal with de-noising by wavelet transform. It was indicated that this method could significantly improve the signal-to-noise ratio (SNR) and effectively filter the noise peaks.3. Under the above conditions, The 14 kinds of dissimilarity set square of maxingshigan tang were measured. The change of chemical constituents of the common decoction and the along decoction were analyzed using wavelet toolbox.
