Objective:The present study investigated the effect of blood loss on tissue-type plasminogen activator(t-PA) in the coagulation and fibrinolysis system in rats. Research and clinical t-PA in older hip fracture mechanism of recessive bleeding.Methods:The rats were randomly divided into three groups(10 animals in each group):control,chronic blood loss groups and actute blood loss groups.After making animal model of anemia, ChenYunLiang reference Shang Cheng and experimental animal model, a method of shear mouse tail blood and preparation of animal models in rats, according to anemia blood loss and electrocardiogram curve,65% of the Hb detection for normal blood, about 4ml, (ECG changes as blood loss rate increase gradually in rats, increase gradually in before the peak, blood loss for 4ml). Anemia is achieved. We detected the content of Hemoglobin (Hb) in blood.Then, we tested t-PA level in plama by ELISA in consective 7 days without any intervation.Results:The t-PA level increased in plasma when blood lossed and Hb decreased,and there is significant difference between control and blood loss groups. In chronic blood loss group,the content of t-PA in plasma increased markedly at first, then decreased gradully and lastly maintained a high level with the increase in blood loss and decrease in Hb.In acute blood loss group,the level of t-PA reached a peak and fell rapidly,then increased slowly,and the range of increase was lower than that of actute blood loss group.Blood lossed can make the plasma t-PA increased, indicating that the blood of recessive t-PA effect more apparent. Whatever the acute or chronic bled blood, stress response t-PA content increased rapidly, will be with the passage of time t-PA will decline, t-PA content that the decline gradually reduce and anticoagulants increased risk of thrombosis. After the first blood group in 2-3 days for turning point, t-PA gradually decreases. After the first blood group Ai 2-5 days t-pa changed after the first blood group A23 days, but slowly began to rise high into the first group, not A, six days, decrease blood began after 2-3 days, cruor-fibrinolysis system activation and balance, such as t-PA, anticoagulant effect in strengthening, lurk bleeding tendency. Blood is 2-3 days after the adjustment cruor-fibrinolysis system, is the key period of thrombosis and bleeding tendency, the active period of clinical attention should be trauma and operation within 2-3 days after cruor-fibrinolysis system, is the prevention and treatment of deep vein thrombosis and recessive bleeding.Conclusions:The level of t-PA increased in plasma with blood lossed.After the wound is 2-3 days, cruor-fibrinolysis system is the key period of adjustment, thrombosis and bleeding tendency of active period.
