Objective:The establishment of experimental rat model of type 2 diabetes, and impose on psychological stress, by comparing the different groups of rats with Thl-type cytokines (IL-2) and Th2-type cytokines (IL-4) and IL-18 in serum of content to explore the psychological stress on immune function in diabetic rats and its related mechanisms.Methods:1,Experimental type 2 diabetic rat model building.60 rats to adapt to a week later, according to the principles of the rats were randomly divided into model group (40 rats) and healthy group (20 rats), building modules using high-sugar high-fat diet plus low-dose repeated intraperitoneal injection of streptozotocin STZ method of establishing experimental rat model of type 2 diabetes.After the end of the experiment by intraperitoneal blood,the model control group and healthy control rats insulin were 26.294±5.928 (IU/ml), and 33.294±2.382 (IU/ml), P= 0.020.Glucagon were 380.607±64.128 (pg/ml), and 296.308±68.759 (pg/ml), P= 0.036.Insulin sensitivity index were-6.021±0.345 and-5.044±0.158, P= 0.000. Consistent with experimental type 2 diabetic rats glucose metabolism characteristics.2,The implementation of psychological stress. From reaching the standard type 2 diabetes mellitus in rats according to the principle of random blood glucose taking 20 as a model group, and were randomly divided into model control group and model stress group. The healthy group rats were randomly divided into healthy control group and healthy stress group. The model stress group and healthy stress group were use of forced cold swim stress and the method of restraint stress on psychological stress.Results:1,Experimental type 2 diabetic rats modeling. The model group have been injected with STZ two days after.Randomly selected rats of model group for fasting plasma glucose 18.739±2.160 (mmol/1), consistent with the standard of experimental model of type 2 diabetes.2,IL-2 rats in each group results. Health stress group and the healthy control group IL-2 levels were 159.504±65.464 (ng/ml) and 111.124±33.348 (ng/ml), model stress group and model control group IL-2 levels were 176.848±61.462 (ng/ ml) and 221.015±48.847 (ng/ml), A factor main effect P= 0.017, B factor main effect P= 0.531.3,IL-4 rats in each group results. Stress group and the healthy control healthy rats, the level of IL-4 were 42.394±9.923 (pg/ml), and 36.781±6.035 (pg/ml), model stress group and model control group, IL-4 levels were 64.528±8.388 (pg/ ml), and 53.317±10.834 (pg/ml), A factor main effect P= 0.023, B factor main effect P= 0.735.4,IL-18 rats in each group results.Health stress group and the healthy control group, levels of IL-18 were 65.514±15.362(ng/ml), and 57.618±13.607 (ng/ml), model stress group and model control group IL-18 levels were96.652±15.501 (ng/ ml), and 77.147±12.235 (ng/ml), A main effect of factor P=0.007, B main effect of factors P= 0.207.5,Model stress group and model control group IL-2, IL-4 and IL-18 comparison. Model stress group and model control group IL-2 levels were 176.848±61.462 (ng/ml), and 221.015±48.847 (ng/ml), P= 0.195; IL-4 levels were 64.528±8.388 (pg/ml), and 53.317±10.834 (pg/ml), P=0.026; IL-18 levels were 96.652±15.501 (ng/ml), and 77.147±12.235(ng/ml), P=0.030.Conclusion:1,Experimental preparation of type 2 diabetic rats may lead to changes in immune function in rats.2,Psychological stress little effect on immune function in healthy rats.3,Psychological stress to experimental rat model of type 2 diabetes, Thl-type cytokines (IL-2) was not obvious, but Th2-type cytokines (IL-4) have significant impact.4,Psychological stress can change the experimental type 2 diabetic rats the level of IL-18.
