Objective To investigate the effects of insulin(INSU) and alendronate(ALEN) on osteoblast-MC3T3-E1 proliferation and apoptosis in high glucose(HG)Methods Osteoblast-MC3T3-El(1×106·mL-1)cells were seeded into the DMEM medium. Then the culured osteoblast-MC3T3-El cells were exposed to high glucose(30mmol·L-1), insulin (10-6mol·L-1) and alendronate (10-7mol·L-1) for 24h, five test groups included high glucose, insulin and alendronate groups (HG, HG+INSU, HG+ALEN, HG+INSU+ALEN) and control group (NG). Cell proliferation were tested by 5-bromo-2'-deoxyuridine incorpration assay, the apoptosis was analysed by flow cytometry.Results Insulin and alendronate may promote expantion of osteoblast-MC3T3-E1 mass as well as decrease apoptosis1 A proliferation of determination results:The results with the experimental group and control group the percentage of absorbance values expressed in each group showed:HG groups, HG+INSU Group, HG+ALEN Group, HG+ INSU+ALEN compared with the control group there was significant difference (p<0.05), and then on each group pairwise comparison, HG+INSU+ALEN group than in HG groups, HG+ INSU Group, HG+ALEN group there was significant difference (p<0.05), and HG+INSU group HG+ALEN group no significant differences between (p> 0.05). Figure 12 osteoblast apoptosis resultsFlow cytometry showed that:in each group showed different degrees of apoptosis, the apoptosis rate after the intervention were to reduce, in high glucose the rate of apoptosis of osteoblasts for the single plus insulin and alendronate sodium were reduced compared with the control group were significantly different (p<0.05), but the HG+INSU+ALEN group than in other groups there were significant differences (p<0.01). Figure 2~Figure 7Conclution Insulin and alendronate may protect osteoblast-MC3T3-E1 in high glucose.
