In recent years, the incidence of breast cancer has gradually increased. The high metastasis leads to significantly low survival rate of patients with breast cancer. Tumor metastasis involves a complex and multistage immune escape. Malignant tumor suppress and destroy the immune system of patients. Radiotherapy, chemotherapy and other treatments all decrease immune function which provides conditions for carcinoma invasion and metastasis. Resveratrol and luteolin have a variety of pharmacological effects, such as scavenging free radicals, antioxidation, antiinflammation and antitumor. Here, we want to explore whether resveratrol and luteolin can inhibit the metastasis of breast cancer.In this study, influence of resveratrol and luteolin on 4T1 breast cancer cells, T cells and B cells was observed in vitro by MTT method. The antimetastatic efficacy in vivo was evaluated by spontaneous lung metastasis model, passive lung metastasis model and the rate of metastasis and recurrence after breast cancer removement. The mechanism was investigated by single cell gel electrophoresis assay and immunity-associated experiment.Both resveratrol and luteolin could inhibit the 4T1 cells growth at dose less than 100μmol/L under normoxia and hypoxia. They could significantly promote T cells proliferation induced by ConA at dose less than 20μmol/L under normoxia and hypoxia. Resveratrol inhibited B cells proliferation induced by LPS. Luteolin showed the same effect under hypoxia and the opposite effect under normoxia.Both resveratrol and luteolin could decline lung tumor node in spontaneous lung metastasis model and passive lung metastasis model. Luteolin could prevent primary breast cancer from growing, and tumor inhibition rate in the prevention group and the treatment group were 41% and 44%, respectively. However, both of them had no effect on the metastasis and recurrence after breast cancer removement. Further study showed that resveratrol and luteolin could protect lymphocytes against H2O2-induced DNA damage. However, they could not enhance immunity- associated response .In conclusion, our results indicated that resveratrol and luteolin could inhibit breast cancer invasion and metastasis by killing tumor cells, enhancing immune function and proventing lymphocytes from DNA damage. But there would be a more complex mechanism of immune modulatory in pathological condition.
