Lung cancer is one kind of malignant tumors with the highest incidence and mortality. The early-stage diagnosis and suitable treatment is important for the patient's to improve their life quality and survival rate. To increase the survival rates of lung cancer patients, we have to solve the problem that we should make the diagnosis earlier. It is expected to make the earlier diagnosis by detecting aberrant methylation of p16,Rassfla,E-cadherin and MGMT genes of lung cancer patients and analyzing the correlation between them. In order to detect the promoter hypermethylation of pi6, Rassfla, E-cadherin and MGMT genes in plasma DNA of NSCLC patients, the plasma DNA was amplified by PCR with methylated-specific and unmethylated-specific primers of p16,Rassfla, E-cadherin and MGMT genes. The frequency of promoter hypermethylation detected in NSCLC patients'plasma for p16,Rassfla,E-cadherin and MGMT genes was 46.8% (22/47)> 42.6%(20/47),31.9(15/47) and 40.4%(19/47), respectively. The methylation analysis with the four genes(p16,Rassfla,E-cadherin and MGMT genes) in plasma free-DNA in this study are obviously improved comparing to the only one gene. The correlation of the promoter hypermethylation of p16,Rassfla,E-cadherin and MGMT genes in plasma was identified. Methylation analysis of p16, Rassfla, E-cadherin and MGMT genes in plasma DNA may be a promising new diagnostic approach to serene putative cancer patients. It will be a very bright future.
