Objective:To investigate the effect of Undaria pinnatifida on anti-tumor activities and to study its mechanism initially for providing a academic basement to research tumor further.To study the contra-Neoplasm metastasis of Undaria pinnatifida fucoidan; to observe the effect of Undaria pinnatifida fucoidan on the growth, tendency and invasion activities of tumour cells.Methods:1.To detect the toxic effect of Undaria pinnatifida fucoidan to SMMC-7721 cells in vitro with MTT.2.Treat the SMMC-7721 with different concentration fucoidan,24h, 48h and 72h.To detect the effect of Undaria pinnatifida fucoidan on inducing tumour cells apoptosis with Annexinâ…¤-FITC/PI3.Treat the SMMC-7721 with different concentration fucoidan,24h, 48h. To detect the expression variation of Survivin and Bcl-2 who are the key factors of apoptosis following Undaria pinnatifida fucoidan treatment with the methods of Western blot, immunocytochemistry and immuno-fluorescence.4. Treat the SMMC-7721 with different concentration fucoidan,24h.To detect the BEL-7402 cells migration following Undaria pinnatifida fucoidan treatment with the experiments of wound-healing model and Boyden chamber model.Results:1.MTT results showed that SMMC-7721 cells were inhibited significantly by Undaria pinnatifida fucoidan, which depend on the drug dosage and treat-time closely. The inhibitory rate of 5-Fu was close to the group of drug 20 mg/ml.2. Flow cytometry showed that the inhibitory rate of tumour cells was correlate with the apoptosis which the fucoidan induced tumour cells. The cell apoptosis increasesd with the time raised. It can be concluded that the tumour cells could be inhibited by Undaria pinnatifida fucoidan, which was positive correlation with treat-time of the fucoidan.3. Immunohistochemistry and immunofluorescence showed that after Undaria pinnatifida fucoidan treatment, the expression of Survivin and Bcl-2 in tumour cells decreased4.Western blot showed that, after Undaria pinnatifida fucoidan treatment, the expression of Survivin and Bcl-2 in tumour cells decreased and leaded to tumour cells apoptosis.5.The experiments of wound-healing model and Boyden chamber model showed that Undaria pinnatifida fucoidan could inhibit BEL-7402 from migrating and reduced tumour cells permeating to peripheral tissue.Conclusion:In our experiment, we can conclude that Undaria pinnatifida fucoidan could inhibit tumour cells from proliferating and promote them to apoptosis, which provide a basement for investigating the biological activities and mechanism of the drug's property of anti-tumour further, and simultaneously, we also confirmed that Undaria pinnatifida fucoidan could inhibit the migration of tumour cells and reduced them permeating to peripheral tissue.
