Objective:The aim of this study was to investigate the expression of MMP-9 and Ki-67 protein in different grades of glioma as well as explore the correlation between them.Method:The expression of MMP-9 and Ki-67 protein in 50 cases of brain glioma were detected with immunohistochemistry and correlation between them were analyzed.10 cases of nomal brain tisse were used as contrast. According to the WHO (Revised 2000) Neuroepithelial tumor separate criteria, we were randomly selected with 20 cases of them are low maglignant group(gradeⅠ-Ⅱ),20 of them are high maglignant group(gradeⅢ-Ⅳ),10 of them are relapse and nomal brain tisse. All the cases did not receive radiotherapy or chemotherapy. All of the immunostained sections were reviewed by double blinded studies. For expression of MMP-9 and Ki-67 protein, ten areas (400×) were selected at random and scored in cases with multiple areas of high intensity that occurred during evaluation of immunostaining. And we calculate the percentage of positive cells. Processing the acquired data with SPSS16.0statistic software.Results:1. Immunohistochemistry were done to evaluate MMP-9 and Ki-67 protein expression, which show brown, respeetively. The Ki-67 Protein expression had a nuclear staining pattern, and the MMP-9 had a cytoplasmic staining in tumor cells.2. The Ki-67 expressed intensity of tumor cells were increase with the histologieal grade up in Gliomas, and application of One-Way ANOVA analysis of variance were low maglignant group 1.27±0.793, high maglignant group 23.440±6.163, relapse group 29.880±18.342. Normal brain tissue group and other groups was a significant difference among them(P<0.05). Low maglignant e group and high maglignant group was a significant difference among them(P=0.000, P<0.05). There was a significant difference between low maglignant group and relapse group(P=0.005, P<0.05). There was no significant difference between high maglignant group and relapse group(P=0.931, P>0.05).3. The MMP-9 expressed intensity of tumor cells were increase with the histologieal grade up in Gliomas, and application of One-Way ANOVA analysis of variance were low maglignant group 23.900±14.083, high maglignant group 46.550±15.385, relapse group 58.560±21.655. Normal brain tissue group and other groups was a significant difference among them(P=0.000, P<0.05). Low maglignant group and high maglignant group was a significant difference among them(P=0.000, P<0.05). There was a significant difference between low maglignant group and relapse group(P=0.003, P<0.05). There was no significant difference between high maglignant group and relapse group(P=0.595, P>0.05).4. There was a positive correlation between the positive expression percentage of MMP-9 and Ki-67 protein in human brain glioma. The differences between the two groups are all have the significance(r=0.695, P=0.000, P<0.05). MMP-9 expression in gliomas is positively correlated with the expression of Ki-67and the malignaney of giiomas.Conclusion:1. The MMP-9 and Ki-67 protein in normal cerebral tissues is negative or weakly expressed.2. In different brain tissues, the rates of the expression of MMP-9 and Ki-67 protein inereased gradually from normal brain tissue to low potential malignancy and high potential malignaney brain glioma. It implied that the expression of MMP-9 and Ki-67 protein correlated with the oceurrence of the glioma.3. The MMP-9 and Ki-67 protein protein are realated to the malignancy and histological differentiation of tumor cell in human brain glioma, can be used as indicators to detection the biological behavior of human glioma.4. MMP-9 expression in glinmas is positively correlated with the expression of Ki-67 and the malignaney of gliomas. The irnrnunohistoehemieal examination of MMP-9 and Ki-67 expression in glioma. which have difference histological grade, should be help for evaluated the tumor cell malignant degree and have significant to demonstrate the mechanism of the carcinogenesis of glioma.
