[Objective]To investigate changes of systemic hematogenic quick innate immunoreactions in patients with acute cerebral infarction(ACI) by nature experimental system in vitro, and approach the capacity of interleukin-8 (IL-8),interleukin-10(IL-10,Duffy antigen /receptor for chemokine (DARC).[Methods]Patients with acute cerebral infarction within 72 hours after onset for the case group and which was the control group with risk factors of atherosclerosis, but never occurred for cerebrovascular disease. Whole blood cells nature experimental tube (0.2ml whole blood cells suspension+0.2ml Escherichia coli suspension inactivated+0.3ml plasma) and whole blood cells nature contrast tube(0.2ml whole blood cells suspension+0.2ml normal saline +0.3ml plasma).Then incubated for 1h at 37℃. The level of IL-8 and IL-10 were measured by ELISA, the level of DARC were measured by FCM with direct fluorescent anti body technique.Then compared between groups.[Result]1.The level of IL-8 was significantly higher in whole blood cells nature contrast group of ACI (154.55±46.83) than it of the control group (29.82±5.31, P<0.01; The level of DARC was higher in whole blood cells nature contrast group of ACI (13.411±1.87) than it of control group (7.34±2.36),P<0.01; But the level of IL-10 was indifference in whole blood cells nature con-trast group of ACI (2.02±1.09) than it of control group (1.47±0.98),P>0.05; Indicating that inflammation reaction of patients with cerebral infarction in the acute phase:The level of IL-8 and DARC highly expressed but IL-10 normal expre-ssion.2. For the case group/the control group:the level of IL-8 was significantly higher in whole blood cells nature experimental group (577.29±73.46)/(135.11±28.26) than it in the whole blood cells nature contrast group (154.55±46.83)/(29.82±5.31),P<0.01; The level of DARC was higher in whole blood cells nature experimental group (16.33±7.35)/(8.91±2.83) than it in the whole blood cells nature contrast group (13.41±1.87)/(7.34±2.36),P<0.05; But the level of IL-10 was indifference in whole blood cells nature experi -mental group (1.82±7.35)/(1.56±0.88) than it in the whole blood cells nature contrast group (2.02±1.09)/(1.47±0.98),P>0.05; Indicating that both for the case group or the control group, Escherichia coli as a strong activator antigen, can rapidly activate the sys-temic hematogenic quick innate immuno-reactions in vitro,modulating the adaptive immunoreactions of white blood cells and the innate immunoreactions of red blood cells.3. The activation rate of Escherichia coli on IL-8 was higher in ACI (5.06±0.83) than it in the control group (3.34±0.82),P<0.05; But the activation rate of Escherichia coli on IL-10 was indifference in ACI (0.44±0.52) than it in the control group (0.56±0.27),P>0.05; The activation rate of Escherichia coli on DARC was indifference in ACI (0.28±0.09) than it in the control group (0.26±0.09),P>0.05; Indicating that the regulation for systemic hematogenic innate immunoreaction to the adaptive immunoreaction was being unba-lanced, IL-8 was significantly value-added, but IL-10 and DARC had no significantly value-added, maybe one of upstream regulatory mechanisms of the inflammation reaction in patients with ACI.[Conclusion]1. Inflammation reaction of patients with cerebral infarction in the acute phase:The level of IL-8 and DARC highly expressed but IL-10 normal expression.2. the regulation for systemic hematogenic innate immunoreaction to the adaptive immunoreaction was being unbalanced, IL-8 was significantly value-added, but IL-10 and DARC had no sign-ificantly value-added, maybe one of upstream regulatory mechanisms of the inflame-mation reaction in patients with ACI.3. Indicating that both for the case group or the control group, Escherichia coli as a strong activator antigen, can rapidly activate the systemic hematogenic quick innate immunoreactions in vitro,modulating the adaptive immunoreactions of white blood cells and the innate immunoreactions of red blood cells.4. The nature experimental system of systemic hematogenic quick innate immunoreactions in vitro is simple, objective, comprehensive and suitable for promotion.
