Object: This experiment, having restraint stress as a model, researched the impact of diffirent stress load on oxidative stress in murine prostate and the adjustment of Bilberry (Vaccinium myrtillus L.) Extract(AN). To investigate the affect of oxidative stress on experimental BPH, the restrait stress was added to the mice with subcutaneous injection of testosterone propionate and the improvement of combination of antioxidant(AN) was observed.Method:Kun Ming (KM) mice loaded with diffirent restraint stress time was observed the changes of oxidative stress in prostate. Animals were randomly divided into 0 h,6h,12h,18h,24 h and 30 h restraint stress time group, The effect of diffirent stress load on prostate of mice were observed in Malondialdehyde (MDA) level and oxygen radical absorbance capacity(ORAC) value in prostate. Mice was subjected to restraint stress time when the status of oxidative stress in prostate reached the maximum and divided into control group,stress group,VitC (200 mg-kg-1),AN (100 mg-kg-1) and AN (200 mg-kg-1). MDA level,glutathione (GSH) content GSH-PX and SOD activities and ORAC were investigated.Restraint stress was combined to experimental BPH modle induced by subcutaneous injection of testosterone propionate(TP) in the male experimental mice. Mice were randomly divided into normal control group, TP group, stress+TP group, CS (200 mg-kg-1)+AN (200 mg-kg-1), CS (100 mg-kg-1)+AN (100 mg-kg-1), AN (200 mg-kg-1), CS (200 mg-kg-1). weight of prostate, Prostate Index, ACP activity in serum,5AR activity in prostate,T level in serum were detemined.for tracing BPH, MDA, ORAC, GSH content and SOD,GSH-PX activity of prostate for oxidative stress in protate.Result: oxidative stress in prostate of mice was significantly induced by 6 h restraint stress, and to the max in 18 h. An oral administration of AN to restrained mice had shown to be decreasing MDA content and increasing ORAC, GSH content, SOD and GSH-PX activities in prostate. oxidative stress in prostate induced by restraint stress obviously aggravated experimental BPH by subcutaneous injection of testosterone propionate(TP). Therapy by CS in combination to antioxidaive AN achieved the prominent effect.Conclusion:These findings suggested that AN may have positive effect towards oxidative stress in murine prostate induced by restraint stress, and combination of AN achieved the best therapeutic effect by scavenging free radical, alleviating oxidative stress.Novel Points:1 For the preventation of disease of prostate, we have restraint stress modle to induce oxidative stress in prostate of mice and investigate oxidative damage of prostate by stress, then improvement of AN on oxidative damage in prostate was observed.2 To observe the impact of oxidative stress by restraint stress on experimental BPH by subcutaneous injection of testosterone propionate in mice, and evaluate the therapeutic effect by CS combination of AN.
