ObjectMouse model building ventilation barriers were observed at different time points, changes in olfactory mucosa of the tissue.MethodsBorn 50 mice were randomly divided into experimental group (20), the control group (30 rats). Experimental mice immediately after birth with a 9-0 suture under the microscope before the left nostril, causing the left front nostril atresia. After 2 weeks,4 weeks,6 weeks,8 weeks, treated mice. HE staining of olfactory mucosa, the thickness of the olfactory epithelium in mice, cell volume changes; and line immunohistochemistry, mice were observed with a microscope in the mature olfactory epithelium olfactory sensory neurons (olfactory receptor neurons, ORNs) and olfactory mucosa histological changes.ResultsNewborn mice (as positive control and negative control), on both sides of the thickness of olfactory epithelium, mature olfactory sensory neurons are similar. Before suturing the left nostril in mice 2 weeks after surgery, compared with the control group the same side, the experimental group slightly thinner olfactory epithelium, OMP-positive cells slightly less.4 weeks was significantly thinner than the olfactory epithelium, OMP-positive cells become less obvious.6 weeks, significant thinning of the olfactory epithelium, OMP positive cells were significantly decreased and dropped to the lowest level, indicating the left mouse at this time before stitching the impact of the nose has reached the maximum.8 weeks, slightly thinner olfactory epithelium, OMP-positive cells recovered slightly less and the level to 2 weeks. ConclusionsEach time point, the experimental group and control group were left nasal olfactory mucosa has histological case differences between the two groups of olfactory epithelium of OMP-positive cells significantly, indicating the left front nostril closure in mice can lead to the corresponding thinner olfactory epithelium, mature reduce the number of olfactory sensory neurons.6 weeks, the experimental group the thickness of the left olfactory epithelium, OMP-positive cells were minimal, suggesting this point, the suture before the nose of the mouse olfactory mucosa and mature olfactory sensory neurons impact has reached the maximum.8 weeks, the thickness of the olfactory epithelium of OMP-positive cells and returned to the basal level at 2 weeks, you may be prompted:In this experimental method, the mouse olfactory sensory neurons in the regeneration cycle is about 6 weeks. In this study, mice were sutured under the microscope before the nose, it can be used as barriers to building ventilation and reliable method of animal models in mice.
