ObjectiveAstrocytes are the most cell types in central nervous system, accounting for 40% in normal adult brain cells, in the provision of central nervous system metabolism and nutritional support and maintain normal function of neurons,regulating synaptic activity have played an important role. Glial fibrillary acidic protein is major intermediate filament protein in mature astrocytes of the brain, that is a major cytoskeletal protein, a large number of GFAP are rich and specific expression in the astrocytes and it is a marked protein in activated astrocytes. After local cerebral ischemia, the changes of GFAP in focal and penumbra area have attracted extensive attention and had in-depth study, In recent years, change of GFAP after ischemia in distant areas also attracted widespread attention. Clinical studies and animal experiments show that the damage is not limited to cerebral infarction local, infarction-related parts that far away from infarction can occur the secondary damage after the acute phase. The purpose of this experiment to observation the distant area of the cerebellum GFAP expression after right middle cerebral artery occlusion in different periods.Methods60 healthy male Wistar rats, Weighing 250-300g, were randomly divided into sham operation 1d group (n=6),3d group (n=6),5d group (n=6),7d group (n=6), 10d group (n=6). After the middle cerebral artery occlusion 1d group (n= 6),3d group (n= 6),5d group (n=6),7d group (n=6), lOd group (n=6). Reference to Zea Longa to prepare rat middle cerebral artery occlusion's suture. Zea Longa score 5 grade 4 points systemand the score at 1-3 into the next experiments. Take four rats in each group to HE stain and immunohistochemistry. Take two rats in each group toelectron microscopy observe. HE observed the tissue morphology of cerebellum, electron microscopy observed ultrastructural changes of cerebellar cortex, immunohisto-chemistry to detect GFAP expression, then statistical analysis, (P<0.05) significant difference.Results1. HE observations show:After the middle cerebral artery occlusion 1d,3d,5d,7d, 10d group compared with the sham-operated group have the normal number of nerve cell, normal from distribution, no significant change.2. Transmission electron microscopy observations show:After the right middle cerebral artery occlusion on the left cerebellum cortex and sham-operated group have no apoptotic cells in 1d group. After 3d group can be seen early apoptotic cells,5d group can be seen late apoptotic cells,7d group can be seen middle apoptotic cells, 10d group can be seen early apoptotic cells.3. GFAP immunohistochemistry:After the middle cerebral artery occlusion 1d,3d group GFAP was positive expressed in astrocytes cytoplasm, yellow-brown, but a smaller cell body, neurite less, a small number of positive expression, and have showed no significant difference with sham operation group (P>0.05). After the middle cerebral artery occlusion 5d,7d, 10d group can see the positive cells increased, the cell body increases, neurite more, the number of positive cells increased significantly, compared with the sham-operated group were significantly different (P<0.05).ConclusionAfter middle cerebral artery occlusion distant parts of the cerebellum medulla showed dynamic changes of GFAP.
