ObjectiveTo investigate the expression of diaphragmatic Muscle Atrophy Factor MAFbx mRNA (Muscle Atrophy F-Box protein) in rats with Acute Lung Injury induced by lipopolysaccharide (LPS).MethodsEighteen Sprague Dawley (SD)rats were alocated randomly into three groups:a control group(n=6), a LPS 4h group(n=6)and a LPS 24h group(n=6). Fresh lung and Diaphragmatic samples were taken at 4h and 24h after 100μg/kg LPS instilled into the trachea of the rats. Normal saline(0.5 ml/kg)was instilled in the control group.pathological changes were observed on the lung tissue,and MAFbx mRNA of diaphragmatic muscle tissue were detected using RT-PCR.Results(1) pulmonary edema,neutrophils and red blood cells invasion into alveolar were found in the experimental group especially in LPS4h. (2) RT-PCR results suggests that the expression of MAFbx mRNA in LPS4h group [2.12±0.22] were remarkably increased compared with the LPS24h group [1.68±0.36]and the control group [1.46±0.21] (p<0.05), but there was no significant difference between LPS24h group and the control group (P> 0.05).Conclusionthe expression of MAFbx mRNA significantly increased indicated that there was diaphragmatic muscle atrophy in rats with ALI.
