ObjectiveEndometrial cancer for female genital mutilation, one of the three malignant tumors, high-fat age of 58 to 61 years old, accounting for 7% of the total number of female cancer, accounting for 20% of female genital malignant tumors 30%, ranking No.4 women's systemic malignancy bit, there is an upward trend in recent years, studies have shown that the growth of tumor cells by the regulation of growth factor. Transforming growth factorβ1(TGF-β1) is an important growth factor with multiple effects, the role of complex, broad participation in a variety of pathological physiological processes in mammals, affect cell proliferation and differentiation in wound healing, embryonic development, cell extracellular matrix formation, bone remodeling, immune regulation, nervous system and tumor genesis and development play a role in such processes. The role of TGF-β1 playing in the begining and during the endomeyrial cancer has not clearly yet. Our preliminary experiments found that from well-differentiated endometrial carcinoma in the poorly differentiated, TGF-β1 showed a gradual downward trend, with the development of endometrial cancer, myometrial invasion and lymph node metastasis, TGF-β1 positive rate was significantly reduced.,suggesting that TGF-β1 and the progress of endometrial cancer was closely negatively correlated, With the development of endometrial cancer, TGF-β1 expression rate is lower.This study investigated the effect of TGF-β1 in proliferation, cell cycle and apoptosis of the endometrialcarcinoma cell line Ishikawa in vitro.MethodsIn this study, endometrial adenocarcinoma Ishikawa cell line to study the object, MTT assay was used to measure the proliferation of cells at different time and different concentration. Flow cytometry and propidium iodide (PI) staining was employed to analysis of different the role of TGF-β1 concentration of Ishikawa cells after 48h cell cycle and apoptosis changes.ResultsMTT showed TGF-β1 inhibited the proliferation of Ishikawa, and it had significant difference at different concentration of TGF-β1(P<0.05). Apoptosis was detected by flow cytometry showed that, TGF-β1 to promote apoptosis in Ishikawa cells, the percentage of apoptosis increased with the concentration of growth (P<0.05); Cell cycle showed that TGF-β1 blocking cell arrest at G1 phase, S phase was significantly decreased compared with the control group, the difference was significant (P<0.05).ConclusionsTGF-β1 may partly inhibit Ishikawa's proliferation, promote its apoptosis and blocked the Ishikawa cell at G1 phase.
