ObjectiveTo discuss the expressions of S100A4 and KISS-1 in bladder urothelium carcinoma and their significance.MethodsWe selected 60 specimens of bladder transitional cell carcinoma(BTCC)as experimental group and 10 specimens of normal bladder tissue as control group.The specimens of the experimental group are all primary BTCC of surgical excision.They have not received radiotherapy,chemotherapy and hormone therapy before operation.The duration of their age is from 20-81.The number of the specimens which have metastasis is 24.And the number of non-metastasis is 36.The 10 specimens of normal bladder tissue of control group are ablated bladder because of ureteric or pyelic TCC.lt has been confirmed that they have not tumor cells after pathological diagnosis.All the specimens have been processed by formalin fixation and mineral waxembedment.We evaluated 60 BTCCs and 10 controls and examined the presence of S100A4 protein and KISS-1,as detected by immunohistochemical streptavidin-perosidase(S-P)method with tumorgrade,clinical stage and lymph node metastasis.Besides,we analyze whether S100A4protein and KISS-1 exist correlation or not.ResultsThe expression of S100A4 protein is negative in normal bladder tissue and the expression of KISS-1 is positive in it.The expression of S100A4 protein was founded in 70%in BTCCs and KISS-1 was 43.3%(P<0.05).The expression rates of S100A4 protein in histological grade I, II and III of BTCC are 31.3%(5/16),68.4%(13/19) and 96.0%(24/25).The expression rates of S100A4 protein in clinical stage Ta-T1 and T2-T4 are 57.1%(16/28) and 81.3%(26/32).And the expression rates of positive and negative lymph mode metastasis group are 87.5%(21/24) and 58.3%(21/36). The expression rates of KISS-1 in histological gradeâ… ,â…¡andâ…¢of BTCC are 81.3% (13/16),47.4%(9/19) and 16.0%(4/25).The expression rates of KISS-1 in clinical stage Ta-T1 and T2-T4 are 64.3%(18/28) and 25.0%(8/32).And the expression rates of positive and negative lymph mode metastasis group are25.0%(6/24) and 55.6% (20/36).Both S100A4 and KISS-1 in BTCC and normal bladder tissue have significant difference.The expression of S100A4 protein and the loss of KISS-1 was positively correlated with histological grade,clinical stage and lymph node metastasis (P<0.05),but not whit age,sex and the size of tumor (P>0.05).The expression of KISS-1 decreases while the expression of S100A4 protein rises.The relation of their expressions is negative.ConclusionsThe expressions of S100A4 protein and loss of KISS-1 are significantly associated whit tumor progression in BTCCs.Both of them can be the predictors of the biological behavior and trend of the patients with bladder transitional cell carcinoma. The expression of S100A4 protein and the expression of KISS-1 in BTCCs have intimate relation.S100A4 protein may decrease the expression of KISS-1.It perhaps affects the occurrence,development and metastasis of bladder carcinoma.
