Relationship Between E-NOS And NF-κ B Expression In Insulin Resistant Rats And Intervenient Effect Of Rosuvastatin

ObjectiveThe model of insulin resistance was made by high-fat diet. To observe the relationship between e-NOS and NF-κB in insulin resistant rats. To approach the intervenient effect of rosuvastatin on the expression of e-NOS and NF-κB in artery endothelial cells, as well as the influence on the insulin resistance.MethodsInsulin resistant rat model was induced by high-fat diet feeding. Thirty-five male Wistar rats were randomly divided two groups feeding either basic ehow(NF group, n=15) or high-fat diet (HF group, n=20) for 10 weeks, then assessed by euglycemic-hyperinsulinemia clamp technique (EHCT). And the rats in HF group were randomly divided into two groups, rosuvastatin intervention group (HFS, n=5) and not rosuvastatin intervention group (HFN, n=5). After 5 weeks intervention treatmeant, serum lipid assay was performed with enzymatic measure, and insulin was determined by enzyme linked immunosorbent assay (ELISA).The expression of eNOS and NF-κB in artery endothelial tissue was tested with Western blotting.Results1,Insulin resistance evaluation:The average glucose infusion rate (GIR) in HF group decreased significantly compared with NF group (P<0.05);The percentage of epididymis adipose mass in body weight, serum triglyceride, TC and LDL-C in HF group increased significantly compared with NF group (P<0.05). Serum HDL-C in HF group decreased significantly compared with NF group (P<0.05);artery endothelial function and inflammation reaction index:artery endothelial function index-expression of eNOS: The expression of eNOS in NF group is strong positive. Compared with NF group, the expression of eNOS in HF group decreased significantly (P<0.05)Artery endothelial inflammation reaction index-expression of NF-κB:There is no or less expression of NF-κB in NF group, while the expression in HF group is strong positive. Compared with NF group, the expression of eNOS in HF group increased significantly (P<0.05)2,The result of rosuvastatin intervention:Serum FINS, triglyceride and TC increase gradually, but ISI decrease in NF group and not rosuvastatin intervention group. That indicates that insulin resistance increase by age.Compared with NF group, Serum FINS, triglyceride and TC increase significantly in rosuvastatin intervention group and not rosuvastatin intervention group, and EAM /BW,TG,TC decreased significantly in rosuvastatin intervention group compared with not rosuvastatin intervention group.Compared with NF group, ISI decrease significantly in rosuvastatin intervention group and not rosuvastatin intervention group, and ISI increase no significantly in rosuvastatin intervention group compared with not rosuvastatin intervention group (P>0.05);Artery endothelial function and inflammation reaction index artery endothelial function index-expression of eNOS:The expression of eNOS is strong positive in NF group, positive or weak positive in rosuvastatin intervention group, but negative or less in not rosuvastatin intervention group. Compared with not rosuvastatin intervention group, the expression of eNOS in rosuvastatin intervention group increased significantly (P<0.05)Artery endothelial inflammation reaction index-expression of NF-κB:The expression of NF-κB is negative or less in NF group, positive or weak positive in rosuvastatin intervention group, but strong positive in not rosuvastatin intervention group. Compared with not rosuvastatin intervention group, the expression of eNOS in rosuvastatin intervention group decreased significantly (P<0.05)3,Correlation analysisThere was significantly negative correlation between GIR and BINS (r=-0.824,P<0.05),and significantly positive correlation between GIR and ISI (r= 0.726,P<0.05) for HF group and NF group before rosuvastatin intervention.After rosuvastatin intervention:For EAM/BW, there was positive correlation with FINS,TG,TC (r=0.673,P<0.05), but significantly negative correlation with ISI (r=-0.418,P<0.05);Correlation between GIR and expression of eNOS:There was significantly positive correlation between GIR and expression of eNOS (r=0.735,P<0.05);Correlation between GIR and expression of NF-κB:There was significantly negative correlation between GIR and expression of NF-κB (r=-0.632,P<0.05);Correlation between expression of eNOS and NF-κB:There was significantly negative correlation between expression of eNOS and NF-κB (r=-0.812,P<0.05).Conclusion1,There were inflammation reactions in the artery endothelium of insulin resistant rat. The expression of eNOS decrease, but the expression of NF-κB increase.2,Rosuvastatin could inhibit the activation of NF-κB, rosuvastatin could active eNOS.
